Objective:To observe the expression of transforming growth factor-β1 (TGFβ1) and collagen in the bronchi and lung tissue of chronic obstructive pulmonary disease (COPD) rat models and their relationship.To evaluate the effects of Aminophylline on the expression of transforming TGFβ1 and collagen in the bronchi and lung tissue.Method:35 Wistar rats were randomly divided into 3 groups, the healthy control group (group A), the COPD model group(group B) and Aminophylline intervention group(group C). Rat COPD model was established by intratracheal instillation of lipopolysaccharide(LPS 200ug/ul) twice and daily exposure to cigarette smoke. Aminophylline intervention group received Aminophylline daily for 7days after COPD model has been established. At the end of five weeks, the pathological features of bronchi and lung tissue were observed by hematoxylin and eosin(HE) stain. The expressions of collagen in the bronchi and lung tissue were measured by Masson stain. The expressions of TGFβ1 and type I collagen,type III collagen in the bronchi and lung tissue were measured by immunohistochemistry with the method of Streptomycin avidin peroxidase. Colorful image analysis system of pathology was used in image analysis. The expressions of TGFβ1,type I collagen,type III collagen and the collagen measured by Masson stain were compared in the three groups.The relationship of collagen and TGFβ1 was also studied.Results:The expression of TGFβ1 in bronchi and lung tissue was (11.75±3.14) % in group B, which was significantly higher than that in group A (2.41±1.16)% (P<0.01). In group C, it was (5.54±2.00)%, which was lower than that in group B (P<0.01), but still higher than that in group A (P<0.01). The expression of type I collagen in bronchi and lung tissue was (14.70±3.76)% in group B, which was significantly higher than that in group A (2.87±1.03)% (P<0.01). In group C,it was (7.36±1.39)%, which was lower than that in group B (P<0.01), but still higher than that in group A (P<0.01). The expression of type III collagen was (17.26±3.81)% in group B, which was significantly higher than that in group A (8.28±3.29)% (P<0.01). In group C,it was (11.37±3.29)%, which was lower than that in group B (P<0.01), and there was no significant difference as compared with that in group A (P>0.05). The expression of collagen in bronchi and lung tissue measured by masson stain was(17.18±5.34)% in group B, which was significantly higher than that in group A (7.42±2.97)% (P<0.01).In group C,it was (12.94±4.03)%, which was higher than that in group A (P<0.05), and lower than that in group B, but there was no significant difference between the two groups (P>0.05).There were statistical positive relationships between the relative contents for the collagen and TGFβ1 in bronchi and lung tissue(r=0.835,0.649,0.692, all P<0.01).Conclusion:The increase depositions of collagen in the bronchi and lung tissue are important pathologic characteristics of airway remodeling in COPD. The expressions of TGFβ1 and collagen in bronchi and lung tissue increase in COPD and the expression of TGFβlis positive relate with the expression of the collagen, which suggests that TGFβ1 may play an important role in the airway remodeling in COPD. Aminophylline decrease the expressions of TGFβ1 and collagen in the bronchi and lung tissue in COPD, which suggests that Aminophylline may be of use in the inhibition of airway remodeling in COPD and may be achieved through the inhibition of TGFβ1. Further studyies are need to identify the main pathophysiology machanism of airway remodeling in COPD and the valid therapy to it.
