Objective To observe the quantity and shape of bone resorption pits and the number of osteoclasts that are induced by different doses of icariin in PMMA cement particles on bone resorption, so as to explore the effects of icariin on this osteoclastic osteolysis, and to evalue the function of it on prevention and treatment of aseptic loosening of prostheses.Methods The familiar protheses wear debris:PMMA Cement Particles are selected as revulsants. Icariin are selected as intervene medicines.30 mice aged 2 monthes are selected. There are five groups.No any disposal is one group. PMMA Cement Particles without fed is one group. Mice fed with 30mg/kg,60mg/kg,120mg/kg with wear debris is respectively one group. PMMA Cement Particles are spread onto their calvarial bones, and then,the difference dosage medicines are fed into them every day. These mice are sacrificed on fourteen day and calvaria are harvested and stained for observed resorption areas and the number of osteoclasts.Results The number of absorption pits of bone slices that are stimulated by wear debris is more than no disposing group(P<0.05),and absorption pits area is also bigger than no disposing group(P<0.05).ICA group have formed smaller absorption pits and area than model group (P< 0.05).Of them, ICA120mg/kg group is most evidence, ICA30mg/kg group being lest. Model group and ICA30mg/kg is no evidence difference respectively(P>0.05)Conclusion PMMA Cement Particles can induce bone absorption and accelerate osteoclasts to generate.ICA are able to hinder the osteoclastic generation and function,so to restrain periprosthetic osteolysis.Therefore,it play an important role in prevention and treatment of artificial joint aseptic loosening.
