Objective: To investigate the effect of combination of preconditioning and postconditioning of different doses of Edaravone on rat kidneys with ischemia-reperfusion injury.Methods: Forty normal male Wistar rats were divided randomly into 5 groups(n=8):sham-operated group(group S),ischemia-reperfusion group(group I),group E1, group E3, group E5 .The rats were anesthetized with intraperitoneal chloral hydrate 300 mg/kg.Bilateral kidneys were exposed through midline incision and bilateral renal pedicels were occluded 45 min with atraumatic mini-clamp then unclamped for 6 h.Rats were treatd before and after ischemia with Edaravone to group E1(1mg/kg), group E3(3mg/kg), group E5(5mg/kg) or saline to group S and group I,administered by tail vein injection.The animals were killed for 6 h of reperfusion.Blood samples were collected from heart for determination of serum creatinine(Cr) and urea nitrogen(BUN) concentrations.Kidney specimens were collected immediately for edtermination of MDA level and SOD activity, the expression of Bcl-2 and Bax by immune-histochemistry assay.The apoptosis in the nephridial tissue was assessed using TUNEL and apoptotic index(AI)was calculated.Results: AfterI/R,the serum Cr and BUN concentrations,the MDA contents, the expression of Bax and the AI were all significantly increased while the SOD activity, the expression of Bcl-2 were siginificantly decreased as compared to the group S(P<0.05). The Cr and BUN concentrations ,the MDA contents, the expression of Bax and the AI were significantly lower while the SOD activity, the expression of Bcl-2 were significantly higher in group E1,E3 and E5 as compered with group I(P<0.05).There were no difference in the indexes among the group E3andE5.Conclusions:1 The combination of preconditioning and postconditioning of different doses of Edaravone can protect the kidney against ischemia-referfusion injury in rats.With the does increase,the effect enhance between 1mg/kg to 3mg/kg.When it reaches 5mg/kg,the effect no more enhance.2 Edaravone can reduce the risk of renal dysfunction and injury by scavenging oxygen free radicals and reducing apoptosis.
