Objective:With the socio-economic development and the process of aging population accelerating,the number of the patients who suffered from high-energy trauma and osteoarthritis increasing, articular cartilage injury and repair has become troubled by the basic research and clinical orthopedic treatment of difficult issues. Therefore, the systemic approach of articular cartilage injury and repair is of great significance. The articular cartilage damage and joint bleeding have some close ties, existing research shows that: whole blood or some of its components only contact with chondrocytes in cartilage cells in a few hours would also cause very serious damage. In addition, some scholars study: A large number of joint bleeding on the articular cartilage of the joint impact than a small amount of bleeding should be obvious.But the composition of blood is abundant, so this experiment is aim to investigate the effect of the rabbit PRP to the knee joint and find out the role of the platelet-rich plasma on the component of articular cartilage, such as water, protein, polysaccharide and collagen,otherwise the main changes and the rules of it will be depicted. Therefore, we designed the following experiment in order to make an institutional explored ?Through autologous PRP re-injection of rabbit articular cartilage of experimental studies, and then an analysis of major changes in the parameters observed indicators, and further explore the PRP re-injection of autologous articular cartilage impact. To provide the research methodology and theoretical basis to study the influence of the others components in whole blood to rabbit articular cartilage.Methods:The 24 New Zealand white rabbits were anesthetized, blood collected from the centrifuge body get PRP, re-injection the right posterior knee joint cavity, left posterior knee as a blank control group, in the third days repeated (dosage and method of op). And then 24 experimental animals were randomly divided into 3 groups, respectively, 1st week, 3rd week, 5th week ,then draw the materials from articular cartilage and blood collection to measure moisture content, collagen content of hydroxyproline, serum MMP-3 and TIMP-1 levels change, at the same time optical observation. All the data using statistical software package SPSS16.0 statistics, information and data to measure the mean±standard deviation (±s) said the difference between the groups using single factor analysis of variance and Q test, P <0.05 considered statistically significant.Results:1 Gross observationGross observation: Normal articular cartilage surface is very smooth, un- iform,shiny, showing the white porcelain. Articular cartilage color slightly gl- oom on experimental side, the surface roughness. The most obvious 1st week after surgery, the 3rd week gradually improved, the 5th week to restore near- normal luster.2 staining2.1 HE stainingSmooth surface of normal articular cartilage, cartilage cells, evenly distr- ibuted and no clustering chondrocytes, tide line of clear and complete. Expe- rimental group cartilage surface is roughness, and there exists small debris,the microscope shows that the cell was inordinate and the image of is mitotic pha- se increased,besides that the cartilage cell clusters together, the tide lines blur.2.2 Alcian Blue stainingNormal articular cartilage chondrocytes surrounding the acid mucopolysa- cc- haridedark blue, pale blue matrix. Experimental group at 1st week after the first color of the surrounding cartilage cells decreased the most visible and obvious, the 3rd week gradually improved, the 5th week to restore near-normal dark blue.2.3 Periodic acid Schiff reaction (PAS) stainingNormal articular cartilage in the cartilage matrix of neutral mucopOlysa- charide was purple, cartilage cells colorless. Experimental group at 1st week after operation decreased cartilage matrix color of the most obvious, the 3rd week gradually improved, the 5th week to restore near-normal purple.3 Changes in moisture contentThe experimental group at 1st week of articular cartilage of the water content (80.67±4.88)% compared with control group (65.86±3.60)% compared significantly different (P <0.05). Experimental group the 3rd week of articular cartilage in the moisture content (72.90±3.69)% compared with control group (65.73±3.72)% difference was significant (P <0.05). Experimental group 5th week of articular cartilage in the moisture content (65.49±2.36)% compared with control group (65.71±2.91)% compared no significant difference (P> 0.05);The moisture content of the experimental group in comparison 1,3,5 weeks were significantly different (P <0.05), the moisture content of the blank control group, the comparison of 1,3,5 weeks did not differ significantly (P>0.05).4 Changes in hydroxyproline content of collagen.The experimental group at 1st week of articular cartilage in the hydroxyproline content (51.6±3.1) mg / L compared with control group (62.8±2.5) mg / L was significant difference (P <0.05). Experimental group in the 3rd weeks of articular cartilage in the hydroxyproline content (62.2±2.9) mg / L compared with control group (63.8±2.8) mg / L showed no difference (P> 0.05). The experimental group in the 5th week of articular cartilage in the hydroxyproline content (63.8±3.1) mg / L compared with control group (63.4±3.0) mg / L showed no difference (P> 0.05). Experimental group, collagen content of hydroxyproline content in group 1st week compared with 3rd, 5th week significantly different (P <0.05), the first 3rd, 5th week showed no difference (P>0.05). The blank control group in 1,3,5 week all comparisons showed no difference(P> 0.05).5 Rabbit serum MMP-3 and TIMP-1 levels change5.1 Experimental group, serum MMP-3 levels at 1st week (7.6±0.6) ug / L with the 3rd week (10.5±1.7) ug / L was significant difference (P <0.05). The 3rd week with the 5th week (7.6±0.7) ug / L was significant difference(P <0.05). 1st week with the 5th week showed no significant difference (P> 0.05).5.2 Experimental group, serum TIMP-1 levels at the 1st week(3.8±0.7) ug / L with the 3rd week (20.4±3.0) ug / L was significant difference(P <0.05). The 3rd week with the 5th week(7.5±0.9) ug / L was significant difference(P <0.05). Week 1 compared with the 5th week significantly different(P <0.05).Conclusion:Re-injection of autologous PRP articular cartilage may have an impact;Re-injection of autologous PRP on the joint impact of moisture and protein polysaccharides more apparently;Re-injection of autologous PRP on collagen content was less affected;Autologous PRP re-injection of serum MMP-3 and TIMP-1 content of the presentation of changes in volatility.
