Objective: To observe the effect of resveratrol to the activity changes of CYP2E1 which is induced by ethanol, and the apoptosis phenomenon of hepatoma carcinoma cell(HepG2) induced by resveratrol.Methods: To establish the experimental model of human liver cancer cells using a common culture with the resveratrol,the HepG2 were cultured in RPMI1640 which is containing 10﹪fetal bovine serum (FBS).The 3 to 6 generaration of cells which is grow well will be take to the experiment.①To observe the effect of resveratrol to the activity changes of CYP2E1 which is induced by ethanol first,experiment group: there are ethanol group(final concentration is 50,100,150mmol/L), resveratrol group(final concentration is 12.5,25,50,100,150μmol/L),and control group . To observe the effect of resveratrol for the activity change of CYP2E1 which is induced by ethanol through RT-PCR and Western blot.②To observe the effect of resveratrol to the cell cycle and the apoptosis of hepatoma carcinoma cell: there are resveratrol group(final concentration is 12.5,25,50,100,150μmol/L),control group without any drugs, the equal volume of DMSO was added to solvent control group. Application of MTT colorimetric assay cell growth inhibition, the apoptotic cell morphology will be observed by AO/EB double staining, flow cytometry laboratory to observe the apoptosis and cell cycle.Results: 1. Compare with the control group, the activity of CYP2E1 is significantly increased can induced by ethanol in HepG2 cells (P<0.01); and the effect of resveratrol in low concentration group(12.5μmol/L) is non difference compare with the ethanol group, but middle and high concentration group(25~150μmol/L) can significantly inhibit this effect of ethanol(P<0.01).2. Compare with the solvent control group, different concentrations of resveratrol all can significantly inhibit the growth of the HepG2 (P<0.01). 3. Compare with the solvent control group, different concentrations of resveratrol all can significantly induce the apoptosis of HepG2 (P<0.01), and the morphological changes is obviously, at the same time the growth of cell is stoped in G1 phase in cell cycle.Conclusions: 1. The activity of CYP2E1 can significantly increased induced by ethanol in HepG2 cells; and the effect of resveratrol in low concentration group(12.5μmol/L) is non difference compare with the ethanol group, but middle and high concentration group(25~150μmol/L) can significantly inhibit this effect of ethanol.2. Low and middle concentration(12.5~100μmol/L) of resveratrol can significantly inhibit the growth of HepG2, influence the cell cycle and induce the apoptosis of it;bue this effect is no longer enhanced when resveratrol reach the high concentration(150μmol/L),show the protect function to cell can be provided by resveratrol in certain extent.
