| Objective: To explore the effect of insulin(INS) administration on the expression of Bcl-2 protein and the apoptosis of corneal histiocytes following rabbit model of corneal alkali burn.Methods: 35 female New Zealand white rabbits were randomly divided into three groups:group A (normal control group, n=5) , group B (saline group, n= 15) and group C (insulin group, n=15). To establishâ…¡degree model of corneal alkali burn, and respectively to inject normal saline and insulin in the subconjunction each day in group B and group C . The corneal samples were remove and fixed at the 1th day, the 7th day and the 14th day postoperative..the apoptosis of corneal histiocytes were detected by pathological examination,electron microscope and transferase-mediated dUTP- digoxigenin nick and labeling assay (TUNEL). the expression and alteration of Bcl-2 protein using immunohistochemisty.Results: Pathological examination and electron microscopy showed group C corneal epithelium, stroma prosthetic situation better than those in group B. The TUNEL positive cells were present slightly in the epithelium of normal comea and not present on stroma and endothelium.A higher number of Bcl-2 expression in the epithelial lining of normal cornea, a lower number of Bcl-2 expression on stroma and endothelium. The TUNEL positive cells of group C was significantly lower than that(P<0.01) in group B, and the expression of Bcl-2 in group C was markedly increased on the 7th day and the 14th day in comparison with group B (P<0.01).Conclusion: Through increasing the expression of apoptosis inhibiting gene Bcl-2, insulin injecting in the subconjunction could inhibit the apoptosis of corneal histiocytes after rabbit corneal alkali burn and promote cornea repair.
