PartⅠ: The intervention in endotracheal tube-associated biofilms and respiratory infections of Pseudomonas aeruginosa by using inhalation of combination erythromycin and ambroxol with ciprofloxacin respectively in rats model【Objective】To research CFU of inhalation of erythromycin and ambroxol combining ciprofloxacin respectively against respiratory infection with endotracheal tube (ETT)-associated biofilm of Pseudomonas aeruginosa.【Methods】Rat model of P. aerudomonsa biofilm infection was established. The bacterial colony count in ETT biofilm and lung tissues were performed after intervention by follow 5 groups: normal saline control as group1, ambroxol as group2, erythromycin as group3, ambroxol combining ciprofloxacin as group4, erythromycin combining ciprofloxacin as group5. Scanning electron microscopy (SEM) on biofilm after 7 days of intervention with above 5 groups respectively.【Results】After 7 days of intervention with above each groups respectively, The bacterial colony count (BCC)from lung tissues(×CFU/ml) from normal saline control as group1, ambroxol as group2, erythromycin as group3, ambroxol combining ciprofloxacin as group4, erythromycin combining ciprofloxacin as group5 respectively, were 139.25±42.01,101.62±40.41,109.62±33.47,22.25±17.31and 57.75±37.82. It were significantly different that the group1,group2 and group3 were compared with the group4 and group5 respectively(P<0.05) ; and it also was significantly different that the group4 was compared with the group5(P<0.05), but not significantly different between the group1,group2 and group3 respectively (P>0.05). BCC from ETT biofilm were as follows (×104CFU/ml): 170.00±48.32,127.62±39.01,133.50±33.68,38.12±19.10and 70.37±35.77 respectively.the results were the same as the lung tissues. The matrixes of the ETT surface was thickest in the group1 by using Scanning Electron Microscopy (SEM). It were less in the group2 and group3, and it were least in the group4 and group5,when comparing with group4 and group5;The group5 was less a little.【Conclusion】the combination ambroxol with ciprofloxacin has significantly effections on ETT-associated biofilm of P. aeruginosa and relevant respiratory infection, which is better than the combination erythromycin with ciprofloxacin.PartⅡ:Pathological change in hung tissues of tube-associated pneumonia after using inhalation of combining erythromycin and ambroxol with ciprofloxacin respectively in rats model【Objective】To search the effect of inhalation of erythromycin and ambroxol combining ciprofloxacin respectively against pneumonia with endotracheal tube (ETT)-associated biofilm of P. aeruginosa.【Methods】Rats model of P. aeruginosa biofilm infection was established. The bacterial colony count from lung tissues were performed after intervention by the follow 5 groups: normal saline control as group1, ambroxol as group2, erythromycin as group3, ambroxol combining ciprofloxacin as group4, erythromycin combining ciprofloxacin as group5.Light microscope on lung tissues after 7 days of intervention with above 5 groups respectively.【Results】After 7 days of intervention with above 5 groups respectively, Observation of lung tissues by using light microscope shows that af ter 7 days of intervention, infection of lung tissues was very severe in the normal saline control, which is hyperemia and consolidation heavy, the infection are still serious in group2 and group3, with infiltration in a lot of inflammatory cells. But the inflammation in lung tissues was not obviously in the group4 and group5 with few of inflammatory cells in lung tissues.【Conclusion】the combination ambroxol with ciprofloxacin was significantly effective on ETT-associated biofilm of P. aeruginosa and relevant respiratory infection after 7 days of intervention, which is better than the combination erythromycin with ciprofloxacin.
