Objective:To investigate the effect of ultrasound-mediated microbubble destruction on the Sertoli cells of rat testis in vitro.Method:Sertoli cells were isolated and cultured from SD rat testis. After ultrasound contrast agent was added into cell suspension, ultrasound was performed for 60 s at the frequency of 1 MHz, and the intensity of 0.5 W/cm2.At the point of 5 min,2,6,12,and 24 h after ultrasound performed,the expression of vimentin was detected by immunohistochemical staining,SOD,MDA in cell were detected and the cells were counted.Result:Sertoli cells were successfully isolated and cultured from SD rat testis. At the point of 5 min,2,6,12,and 24 h after Ultrasound-mediated microbubble performed the expression of vimentin was: 0.141±0.009,0.149±0.006,0.218±0.007,0.761±0.012,0.847±0.011,the expr--ession of vimentin was significant deviation compared to control group and ultrasound group (P<0.05).the expression of vimentin was no significant deviation between control group and ultrasound group (P>0.05).SOD,MDA in cell at the point of 5 min were:9.15±0.17,11.37±0.48,2 h were 13.25±0.56,12.54±0.36,6 h were 14.05±0.28,13.79±0.62,12 h were9.47±0.12,14.33±0.63, there were significant deviation compared to respectively control group.At 24 h were 3.75±0.48,16.02±0.33,and no significant deviation compared to respectively control group.There were no significant deviation about the survival rate of the cell in control group,ultrasound group and ultrasound-mediated microbubble group at the point 24h,48h (P>0.05). So ultrasound-mediated microbubble destuction decreased the expression of vimentin, increased MDA and decreased SOD at earlier time,but at later time (24 h),MDA became normal,SOD increased and had no effect on Sertoli cells'survival.Conclusion:Ultrasound-mediated microbubble destruction can cause damage on structure and function of Sertoli cells reversibly,which can provide a new method for the study of male contraception.
