Objective :To explore the relationship between mRNA and protein expressions of smac,XIAP and caspase-3 with tumorigenesis and invasiveness of pituitary adenoma .Methods: The protein expressions of smac,XIAP and caspase-3 were detected by immunohistochemical analysis and western blot ,and their mRNA expressions were detected by reverse transcription polymerase chain reaction(RT-PCR)in 59 specimens of pituitary adenoma that was classified into invasive and non-invasive according to synthetic classification.Results: 31 specimens of pituitary adenoma were invasive and 28 were non-invasive.The protein and mRNA expressions of XIAP was significantly higher in invasive pituitary adenoma than that in non-invasive.The protein and mRNA expressions of smac and caspase-3 were significantly lower in invasive pituitary adenoma than that in non-invasive,the texts were statistically significant . The analysis of correlation showed that the positive correlation exists between mRNA and protein expressions of smac and caspase-3 in invasive pituitary adenoma (P<0.01),the Negative correlation exists between mRNA and protein expressions of the former two and XIAP (both P<0.01).There were no positive correlation among the three in non-invasive pituitary adenomas (p>0.01).Conclusion: High expression of XIAP and low expressions of caspase-3 and smac may play important roles in the tumorigenesis and invasiveness of pituitary adenoma,and they can be useful molecular biological markers in determining invasive behavior of pituitary adenoma.
