【Objective】Use of autologous stem cell transplantation in patients with hematologic malignanciesperipheral blood mononuclear cells cultured dendritic cells/ cytokine induced killer cells.Observation of DC/CIK cell proliferation. Application of DC/CIK cellualr immunotherapytherapy with autologous hematopoietic stem cell transplantation in patients with hematologicmalignancies, adverse reactions were observed。【Method】Peripheral blood mononuclear cells from autologous stem cell transplantation completeremission of hematologic malignancies patients, cultured in vitro were amplified by DC and CIK.Observed DC and CIK cell morphology use of inverted microscope . Underwent training processthe relevant pathogen (bacteria, fungi) monitoring. Flow cytometric analysis of DC/CIK cellphenotype.Clinical in vitro induction of DC/CIK by flow cytometry after identification and pathogendetection negative, cell reinfusion by subcutaneous(bilateral neck, armpit and groin) andintravenous injection to patients. Transfusion during and after transfusion observed clinicaladverse events and to monitor the patients blood, urine routine, liver and kidney functionenzymes, and electrocardiogram changes.【Results】DC cultured for 2 days beginning larger cell volume; 3 days cells began to appear deformed,elongated fusiform shape, small dendritic and radial and other irregular shape, some cells beganto suspension, adherent cells were gathered into groups; cultured for 5 days, significantlyincreased colony, more adherent cells in light state; cultured for 7 days most of the cells began tosuspend, irregular shape, a large number of cells with burr-like protrusions, irregular for a typicalDC morphology, some cells were round, star and dendritic-like protrusions. Induced maturationof DC by flow cytometry showed that CD80, CD83, CD86, CD1a positive cells such as rateincreased substantially. CIK culture doubling around 5 days, cell volume increased significantly,showing mother cell-based, and cluster growth, with the cell culture time, the rate of CIK in theCD3 + CD56 +, CD3 + CD8 + double positive cells also significantly increased .In the DC / CIK cell immunotherapy process, all patients in good physical , the course oftreatment and after treatment, some patients had low fever, fatigue, subcutaneous injection site adverse reactions such as mild pain, low fever duration in about 2-6 hours and can subside ontheir own, there were no life-threatening complications and significantly side effects. Monitoringpatients after treatment, blood routine, liver and kidney function, myocardial enzymes and ECGshowed no abnormalities.【Conclusio】Autologous stem cell transplantation in patients with peripheral blood mononuclear cellson DC/CIK cell culture is not affected. Culture methods used in this study can get strongproliferation, cell phenotype associated with higher DC/CIK. DC/CIK cells for clinicalimmunotherapy. DC/CIK cellualr immunotherapy with autologous hematopoietic stem celltransplantation in patients with complete remission of the hematologic malignancies, good safetyand less adverse reactions。...
