Objective:To investigate the role of recombinant human interleukin-10(rhIL-10) in rabbit anti skin graft rejection and its influence on cytokines. Methods:As a receptor of 18 rabbits,18 rabbits as a donor to establish animal models for rabbit skin grafts. the experiment is divided into 6 group:①group:rhIL-10(5μg/kg);②group:rhIL-10(10μg/kg);③group:CsA(5mg/kg);④group:CsA(10mg/kg);⑤group:rhIL-10(5μg/kg)+CsA(5mg/kg);⑥group:received the dose of 0.85% sodium chloride. Observed 28 days after skin transplantation in rabbits in general,the time of skin graft rejection and graft survival time of skin; Preoperative 1d, after 1d,4d,7d,14d,21d and 28d blood serum was separated using ELISA, observed IL-2,IL-4,IL-10,TNF-αand IFN-γchanges in the transplanted tissue. Results:1. Receptor in rabbit skin graft rejection time①,②group were 6.67d and 9.00d,③,④,⑤group were 12.00d,19.67d,16.16d and⑥group was 4.33d. compared with⑥group,①group of skin graft rejection time delayed, but no difference. the time of skin graft rejection in②,③,④,⑤group were delayed, there is difference(p<0.05).2. The mean survival time of receptor in rabbit:①,②group were 13.33d and 16.00d;③,④group were 15.33d and 21.33d;⑤group was19.00d;⑥group was 7.67d.①,②,③,④,⑤group compared⑥group respectively, between the two groups, each has a statistically significant (P<0.05);①,②group were respectively compared with④,⑤group, each has a statistically significant differences (P<0.05).3. The results of IL-2 Test show that:after operation all groups elevated, in which the most obvious was⑥group. in 4-7day, the level arrived their peak, and then show decline, but still higher than other groups, and compared with②,③,④,⑤group, there were significant differences(P<0.05).①group compared with⑤group, there were significant differences(P<0.05).4.The results of TNF-a Test show that:after 1d, compared with the other groups, the expression level of⑥group increased more rapidly, the peak reached in 4d,and then slow decline, but still higher than other groups, compared with the others, there were significant differences (P <0.01). there was no significant difference between①and②group (P> 0.05).5. The results of IFN-y Test show that:After 1day, compared with other groups, the expression level of⑥group increased more rapidly, the peak reached in 4day,and then slow decline, but still higher than other groups. compared with the others, there were significant differences (P<0.05); there was no significant difference between①and②group (P> 0.05).6. The results of IL-4Test show that:In addition to⑥group, the expression level of other groups have slowly increased, and in 4 day, they increased more obvious, which④group and⑤group reached their peak in 7 day respectively,①and②group reached their peak in 14 day,⑥group compared with other groups, there were significant differences(P<0.01), there was significant difference between⑤group and①,②,③group(P<0.05). there was no significant difference between①and②group (P> 0.05).7. the results of IL-10 Test show that:after 1d,⑤group increased faster than other groups, In4d, and then declined at a comparatively low level, Other groups were increased in different degrees, of which the most obvious was②group, in7d after surgery, they arrived their peak; there were significant differences(P<0.05) compared with④,⑤,⑥group; there was no significant difference between①and②group (P> 0.05). Conclusion:rhIL-10 can inhibit the rabbit skin graft rejection, rejection reactions can be so delayed, and prolong graft survival time; indicating that rhIL-10 can inhibit the rabbit skin graft rejection by reduce the expression of IL-2,TNF-α,IFN-γ; study also shows that rhIL-10 combinat with CsA can reduce CsA toxi effects, rhIL-10 and CsA have synergistic effect what is more, they can enhance the effect of anti-transplant rejection.
