Purpose:To investigate the regulation of estrogen on duodenal mucosal bicarbonate transport protein in mice, then elucidate the cellular mechanisms whereby estrogen increases duodenal mucosal bicarbonate secretion and protect gastroduodenal mucosa against damage, and further not only provide theory basis for the prevention and therapy of gastroduodenal mucosal damage, but also contribute to exploring new therapeutic methods for gastroduodenal mucosal damage.Methods:Female C57BL/6J mice with the age of 4 weeks were randomly divided into ovariectomy, shame operate, and normal groups. Vaginal smear was made to determine if ovary was successfully extirpated after ovariectomy. Duodenal mucosae were taken at 3 days and 1 week after ovariectomy. The mRNA changes of duodenal mucosal bicarbonate transport protein (CFTR,SLC26A3,SLC26A6) were determined by using RT-PCR.Results:The cyclic change of vaginal smear in mice with ovariectomy disappeared at 3 days after ovariectomy, presenting numerous leukocytes and keeping in diestrus. But the mice in the shame operate and normal groups had cyclic changes in vaginal smear. The results demonstrated that ovariectomy was successfully performed. The results in RT-PCR showed that mRNA expressions of CFTR,SLC26A3,SLC26A6 in shame operate group had no significant changes at 3 days and 1 week after ovariectomy compared with normal groups (P>0.05). However, in ovariectomy group, the mRNA expressions of CFTR and SLC26A3 had no significant changes at 3 days after ovariectomy (P>0.05), but began to low at 1 week after ovariectomy compared with normal group (P<0.05). And the mRNA expressions of SLC26A6 decreased at 3 days and 1 week after ovariectomy compared with normal group(P<0.05).Conclusion:Estrogen regulated the mRNA expression of duodenal mucosal bicarbonate transport protein, this contributes to elucidating the mechanism whereby estrogen affects duodenal mucosal bicarbonate secretion. The change of mRNA expression of SLC26A6 was faster than the change of CFTR and SLC26A3 in ovariectomy group.
