Objective The study will give a detailed analysis on the gene polymorphism of drug metabolizing enzymes of CYP2D6 in the patients with renal inadequacy in Guizhou through studying the genotype frequencies of CYP2D6*10 in kidney diseases patients and the relation between the gene and renal inadequacy.The study will provide clini-cal drug analysis of individual genetic polymorphisms for clinical rational drug use and provide a scientific basis for the individual dose so as to improve effectiveness and reduce side effects. Methods From March to October in 2009,44 patients with a long-term treatment of hemodialysis from Guizhou Provincial People's Hospital, who were diagnosed renal inadequacy,and 59 kidney diseases patients with serum creatin-ine below 451μmol/Lwere selected to be our research aims.with the patients'con-sents,2ml blood samples from each patient were collected, In the same time, the gen-der,the ethnicity, the serum creatinine, the diagnosis and other general information of each patient were selected. The DNA was extracted by polymerase chain reaction (PCR)amplification of CYP2D6*10 with five primers in a single tube,and detected the gene by the technology of agarose gel electrophoresis in the gel electrophoresis detection imaging device. According to the result of the genotype frequencies of CYP 2D6* 10 in patients with nephropathy in Guizhou Province, the fact wanted to be known that whether the genotype frequencies met the Hardy-Wenberg balance, and whether the genotype frequencies of kidney diseases patients were significantly diffe-rent compared with the normal people.Results①In patients with early renal inade-quacy the allele homozygote TT of CYP2D6*10 is less than heterozygotes CT and is also less than wild-type homozygous CC, the frequencies of TT, CT and CC are 0.305, 0.373 and 0.322 respectively. The gene frequencies of allele T and C are 0.4915 and 0.5085 respectively.②In patients with end-stage renal inadequacy the allele homozy-gote TT of CYP2D6*10 is less than heterozygotes CT and is also less than wild-type homozygous CC,the gene frequencies of TT, CT-and CC are 0.296,0.386 and 0.318 respectively. The gene frequencies of allele T and C are 0.489 and 0.511 respectively.③The differences of genotype and allele frequencies are no significant between the early renal inadequacy group and the end-stage renal inadequacy group(P>0.05).④The differences of gene frequency per person and each genotype of CYP2D6*10 are no significance between the early renal inadequacy group and the end-stage renal in-adequacy group (P>0.05)⑤The differences of kidney diseases·patients'allele fre-quency of CYP2D6*10 are no significance compared with the normal people.Con-clusion (1) the gene frequency per person of CYP2D6*10 and gene frequency per person of each genotype of CYP2D6*10 between the early renal inadequacy group and the end-stage renal inadequacy group are no difference. (2)the genotype and allele frequencies of CYP2D6*10 between the early renal inadequacy group and end-stage renal inadequacy group are no difference. (3) The results show that the genetic poly- morphism of CYP2D6*10 in the development of renal inadequacy have no influence. (4) The study find that allele frequency of CYP2D6*10 is 49.5%, and the gene fre-quency from China Miao nationality is 47.15%, close to 51%among the Han nationa-lity.
