| Abstract:Objective:To study the destruction of endothelial cells and the effective of protection of Losartan on endothelial cell during cardiopulmonary bypass(CPB) in dogs.Methods:Twelve adult hybrid health dogs (12 18kg)were randomly devided into control group(group C, n=6)and Losartan group(group S,n=6),the dogs were anesthesitized with 2.5% sodium pentobarbitone (25mg.kg-1) by intraperitoneal injection, mean artery pressure(MAP) and centre venous pressure (CVP) were measured via femoral artery and femoral vein and simulated the model of CPB in both groups. Losartan 10mg.kg-1 was administered intravenously before CPB, in group S, while it was treated with only physiologica salt solution in group C, The levels of Angiotoninâ…¡(Angâ…¡), Von Willebrand's factor (vWF),Malondialdehyde (MDA) were measured befored CPB(Ti,baseline),45min after cross-clamping(T2),30 min(T3) and 60 min(T4) after aortic unclamping in both groups.femoral artery tissues were obtained before CPB(T1,baseline),45minafter cross-clamping(T2),60 min after aortic unclamping(T3) for microscope examination.Results:There was no significant difference in the levels of Angâ…¡,MDA,vWF before CPB in both groups. The levels of were significantly higher than those before CPB at various time points (p<0.05) in both groups,The levels of MDA and vWF in group S were significantly lower than those in group C at the various time points 45min after CPB(p<0.05),but levels of Angâ…¡has no obviously difference in both groups at all times (p>0.05),the vasclur endothelium was more damaged in group C than that in group S afterCPB. Conclusion:The endothelial cell can be activated and injured by CPB and Losartan might have protective effect on endothelial cell during CPB in dogs. |
