ObjectChenpi and Huoxiang belong to aromatic drugs, the former inclines to liqi, and the latter inclines to eliminate damp. in the process of Huoxiang Zhengqi preparation's production, hesperidin is regarded as the quality monitoring index of Chenpi, patchouli oil is a substance for Huoxiang in which patchouli alcohol is regarded as the quality monitoring index. However, the absorption characteristic of them in alimentary canal and the differences of pharmacologic action between different extractives have no report up to now and deserve being studied. We study the two drugs' characteristics of intestinal absorption and pharmacologic action on gastrointestinal tract via investigating the absorbing characteristics of hesperidin and patchouli and the effects of different extractives from Chenpi and Huoxiang on Limb Ischaemia-Reperfusion (LIR) model rat's gastrointestinal motility and the level of relational gastrointestinal hormones, aiming to provide reliable proof for the research and development of Huoxiang Zhengqi preparation and reasonable exploition of traditional Chinese medicine.Methods1. To investigate the best preparation method of Chenpi and Huoxiang by means of which hesperidin and patchouli alcohol has a high recovery ratio in the decoction via studing the factors of the proportion of drug to water, decoction and concentration method, final concentration of drug.2. To study the the absorbing characteristics of hesperidin and patchouli in rat's small intestine and define the main absorbing place and absorbing mechanism.3. To create gastrointestinal dyskinesia model by LIR and set up groups of different extractives from Chenpi and Huoxiang.4. To compare the pharmacologic actions by studying the changes of gastric emptying and small intestinal propulsive function of different extractives group.5. To measure the level of gastrointestinal hormones in rat's colon mucosa of different groups including substance P (SP) and vasoactive intestinal peptide (VIP) by radioimmunoassay and demonstrate the mechanism of regulative effect of different extractives on gastrointestinal motility.Results1. The best preparation method of Chenpi by means of which hesperidin has a high recovery ratio in the decoction:(1) The proportion of drug to water:1:10; (2) Decocting method:Decocting under 80℃lasting 20min. (3) Concentration method:boil-blowoff lasting 2 min-boil, again and again. (4) Final concentration of drug:2g/mL. We didn't detect patchouli alcohol in the decoction prepared in the same several methods as hesperidin.2. The experimental results of everted sac technique showed that the main absorbing place in rat's small intestine is jejunum and the absorbing mechanism is passive diffusion; in the course of the experiment, we didn't detect patchouli alcohol in the enchyma of any intestinal segment.3. The Gastrointestinal dyskinesia model was created successfully by LIR. In comparison with that in the normal controls, the gastric emptying and small intestinal propulsive function of the model rats decreased obviously with the level of SP decreased and the level of VIP increased obviously in the colon mucosa.4. In the aspect of improving the LIR model rats' gastric emptying function and small intestinal propulsive function, Chenpi water extractives had better effect than Hesperidin, and Huoxiang water extractives plus patchouli oil had better effect than itself while Patchouli alcohol had no effect. There were significant differences between the group of Chenpi water extractives and Hesperidin, between the group of Huoxiang water extractives and Huoxiang water extractives plus patchouli oil.5. In the aspect of regulating the level of SP and VIP, Chenpi water extractives had better effect than Hesperidin, and Huoxiang water extractives plus patchouli oil had better effect than itself while Patchouli alcohol had no effect. There were significant differences between the group of Chenpi water extractives and Hesperidin, between the group of Huoxiang water extractives and Huoxiang water extractives plus patchouli oil.Conclusion In the aspect of pharmacokinetics, we studied the intestinal absorption characteristics of hesperidin and patchouli alcohol, and defined the main absorbing place and absorbing mechanism in the small intestine; In the aspect of pharmacodynamics, we investigated the effects of different extractives from Chenpi and Huoxiang on LIR model rat's gastrointestinal motility and the level of relational gastrointestinal hormones, and provided reliable proof for the research and development of Huoxiang Zhengqi preparation and reasonable exploitation of traditional Chinese medicine.
