Background:Hemostasis played very important role on wound healing and surgical operation. Recently, laser was applied widely in the field of surgical operation, which showed good hemostatic effect. Laser's hemostatic effect was only compared in individual surgical operation, but not researched in the field of wound bleeding, and yet there was not special laser instrument to stop wound bleeding. At present, the laser hematischesis technology was still only limited in the surgical operation or other clinical treatments. Laser showed good hemostatic effect on surgical operation, which also illustrated a good prospect of application in the field of wound hemostasis.Purpose:Through animal experiments to observe the hemostatic effect on local irradiation wound by 532nm laser, explore new methods to stop wound bleeding.Materials and method:(1) The traditional pressure group was used as the control group, and 5W 532nm laser irradiation group was used as the experimental group.Ten healthy, grown, female New Zealand Rabbits weighing (2.5-3.0) kg were selected. Control experiment was carried out with 10 rabbit. A wound of cutting center artery and vena was made on every ear back of each rabbit, which was (0.6-0.8) cm in length,5cm in distance from ear tip. The left ear group was named as tradition group, which was stanched by traditional pressure methods. The right ear group was named as 5W laser irradiation group, which was irradiated by 5W 532nm laser and then pressed to stanch. Hemostatic time was tested. Hemoglutination analysis experiment was performed with heart blood, Prothrombin time (PT), Activated Partial Thromboplastin time (APTT) and Platelet (PLT) number were measured. The wounds were biopsied separately and specimens were observed after HE staining under microscope after 3d,7d, 10d,20d and 30d. (2) 5W 532nm laser irradiation group was used as the control group, and 10W 532nm laser irradiation group was used as the experimental group.Eight healthy, grown, female New Zealand Rabbits weighing (2.5-3.0) kg were selected. The right ear group was named as 10W laser irradiation group, which was irradiated by 10W 532nm laser and then pressed to stanch. Hemostatic time was tested. The wounds were biopsied separately and specimens were observed after HE staining under microscope after 3d,7d, 10d and 20d.Results:(1) The traditional pressure group was used as the control group, and 5W 532nm laser irradiation group was used as the experience group.Hemostatic time showed that the average hemostatic time of tradition group was 306.81 s,and the average hemostatic time of 5W laser irradiation group was 73.55s. The average hemostatic time of 5W laser irradiation group was significantly shorter, there was statistical difference (P<0.05). The results of hemoglutination analysis experiment showed that the average PT of tradition group was 20.77s,and the average PT of 5W laser irradiation group was 21.68s. The average PT of 5W laser irradiation group was longer, there was statistical difference (P<0.05); the average APTT of tradition group was38.09s, and the average APTT of 5W laser irradiation group was 36.61s. The average APTT of 5W laser irradiation group was shorter, there was statistical difference(P<0.05); the average PLT numer of tradition group was 3.52×1011, and the average PLT numer of 5W laser irradiation group was 3.63x1011. The average PLT numer of 5W laser irradiation group was increased, there was statistical difference (P<0.05). The HE staining results showed that 5W laser irradiation increased the tissue damage and prolonged the healing time.(2) 5W 532nm laser irradiation group was used as the control group, and 10W 532nm laser irradiation group was used as the experimental group. Hemostatic time showed that the average hemostatic time of 5W laser irradiation group was 70.36s, and the average hemostatic time of 10W laser irradiation group was 52.82s. The average hemostatic time of 10W laser irradiation group was shorter, there was statistical difference (P<0.05). Compared with the 5W laser irradiation, the 10W laser irradiation made the tissue damage worse, but the HE staining results showed that there was not significant difference between 5W laser irradiation and 10W laser irradiation.Conclusions:Laser irradiation can effectively promote wound hematischesis, but aggravate the tissue damage and prolong the healing time.Laser irradiation on wound has certain influence on PT, APTT and the PLT numer of mechanism of body-wide hemoglutination. It is worth carrying on research that the laser affects the clotting mechanism.
