ObjectiveBy studying electricity for theβ-amyloid protein 1-40(β-amyloid 1-40, Aβ1-40) induced by Alzheimer's disease (Alzheimer's disease, AD) model of cognitive dysfunction in rats, the hippocampus leukocyte inflammatory cytokine interleukin (IL-1β, IL-6) and other aspects of the generation protective effect of inflammatory and immune from the point of view to explore the neuroprotective effect of electro-acupuncture mechanism for acupuncture in the clinical treatment of AD and provide experimental evidence.MethodsOf 48 rats were randomly divided into four groups:normal group 12 animals,12 sham-operated group, model group 12 and the electro-acupuncture group 12. Model group and the electro-acupuncture group were treated with bilateral hippocampal microinjection of Aβ-40 of the state pool 1μ1 established rat model of AD, sham-operated group rats with bilateral hippocampal injection of normal saline. Morris water maze test with rats in each group of learning and memory ability, immunohistochemical determination of the hippocampus of inflammatory cytokines IL-1β, IL-6 expression.Results1.Morris water maze test:(1) place navigation test, with the increase in the number trained, each progressively shorter escape latency in rats, indicating a learning platform is looking for memory processes. However, rats in each group to find the platform for learning and memory abilities showed significant differences. The model group escape latency was the longest, compared with the normal group there was a significant difference (P<0.01), shows model of learning and memory in rats a reduced ability to modeling success. Electro-acupuncture group and model group, escape latency was significantly shorter after withdrawal of the original platform, platform quadrant swim time was prolonged, across the number of the original platform location is also noticeably increased, and the two groups had significant difference (P<0.05). Tip of electroacupuncture treatment on learning and memory in AD rats have some improvement. The normal'group and sham operation group showed no significant difference (P> 0.05), prompt surgical injury in rats did not have a significant learning and memory effects. (2) space exploration experiments, the model group in the original platform quadrant swim time and through the location of the original platform, the number of times higher than those in normal group and sham operation group was significantly reduced (both P<0.01); electro-acupuncture group of rats in the original platform, quadrant swim time and across the former platform location significantly more than the number of model group, There was a significant difference (P<0.05).2. Hippocampal area of inflammatory cytokines IL-1β, IL-6 immunohistochemical staining resultsTest results showed that IL-1β, IL-6 in the normal group and sham group rats were less positive cells in the hippocampus, in comparison with no significant difference. The model group compared with control group, IL-1β, IL-6 in the model group, positive cells in hippocampus of rats increased significantly, positive cells show brown cytoplasm, glial cells and neurons are expressed in the two groups have more significant difference (P<0.01), showed that injection of rat brain targetingβ-amyloid protein 1-40 (Aβ1-40) may promote increased secretion of inflammatory factors, resulting in inflammatory response; electro-acupuncture group compared with the model group, inflammation factor IL-1β, IL-6 positive cells were significantly reduced, with significant difference (P<0.05), prompts electro-acupuncture stimulation can reduce the secretion of inflammatory factors, electro-acupuncture effective treatment.Conclusion1. This subject of study results show that the hippocampus of rats injected through the Aggregation of Aβ1-40 to replicate Alzheimer's animal model is a success.2. Morris water maze test showed that electro-acupuncture can improve the AD model rats in learning and memory impairment.3. Electroacupuncture on AD pathological changes in rat brain has to improve the role of AD can reduce the loss of hippocampal neurons in the phenomenon of inhibition model of rat hippocampal microglia activation and inflammatory cytokine IL-1β, IL-6 over-expression, thereby reducing the inflammatory reaction in brain tissue, thus against the AD.
