Objective To observe the influence of electro-acupuncture on the expression level of SYN and PSD-95 of rats with Alzheimer's Disease, and compare the different effect between electro-acupuncture of different frequency. Try to explore the mechanism of electro-acupuncture treating the Alzheimer's Disease from the standpoint of synaptic plasticity.Methods Select 60 qualified rat by Morris water maze test, and divide them randomly into six groups,a normal group, a model group, a sham operation group, a 2Hz electro-acupuncture group, a 30 Hz electro-acupuncture group and a 50 Hz electro-acupuncture group,10 rats in each one. Use injection of A?1-42 to prepare the model of AD. Acupuncture was applied at “baihui”and“ shenshu ”,the electro-acupuncture groups were provided with electro-acupuncture of 2Hz,30 Hz,50Hz in frequency, once a day, seven days as a treatment course, take a day for rest during the two courses, the treatment include two courses. The expression of SYN protein and PSD-95 protein was determined by Western blot and immunohistochemistry.Results1. Morris water maze test(1) Place navigation test: the model group's escape latency is the longest, exist significant difference with the normal group(P<0.01).Compared with the model group, the escape latency of the three electro-acupuncture groups become shorter obviously(P<0.01).Meanwhile,the escape latency of the 50 Hz electro-acupuncture group is much shorter than the others(P<0.01), there exist no apparent discrepancy between group 2Hz and 30Hz(P>0.05). There also exist no significant difference between the normal group and the sham operation group(P>0.05).(2) Space exploration test: Compared with the normal group,it cost the rats of the model group much more time to swim across the original platform,and the frequency of their acrossing the original platform decreased(P<0.01). Compared with the model group, the rats of the three electro-acupuncture groups spent less time to swim across the original platform(P<0.01), and the frequency they passed through the original platform increased(P<0.01).The time and the frequency of the 50 Hz group shows significant difference with the 2Hz and the 30 group(P<0.01), but there isn't exist any significant difference between 2Hz and30 Hz group(P>0.05).There also exist no significant difference between the normal group and the sham operation group(P>0.05).2. the test result of SYN and PSD-95Compared with the normal group, the expression of SYN and PSD-95 in the model group reduced distinctly(P<0.01). There exist no apparent discrepancy between the normal group and the sham operation group(P>0.01).Compared with the model group, the expression of SYN and PSD-95 in the three electro-acupuncture groups increased obviously(P<0.01), and the higher the frequency of the electro-acupuncture the higher the expression increased,the difference among the groups is obvious(P<0.05,P<0.01).Conclusion1. The model that made by the injection of A?1-42 is success.2. The result of Morris water maze shows that electro-acupuncture can improve the AD model rats' ability of study and memory.3. The result of the Western blot and immunohistochemistry test show that electro-acupuncture can effectively improve the expression of SYN and PSD-95 protein, and enhance the plasticity of synapsis, that may be the possible mechanism that electro-acupuncture work on Alzheimer's disease.4.The effect of electro-acupuncture is related to the frequency, the50 Hz frequency electro-acupuncture has the best effect, the 30Hz's effect is worse, the 2Hz's effect is the worst.
