Objectives:Breast cancer is the highest incidence rate malignant tumor in feminine disease. Its occurrence and development is closely related to abnormal expression of proteins.This study was designed to investigate the expression and relationship of B7-H1 and PD-1 and IL-10 protein in normal breast and benign lesional and breast cancer, and their correlation with clincopathologic features, for provide evidence for breast cancer of diagnosis and prognostic evaluation.Methods:283 paraffin-embedded specimens were collected , including 267 cases of invasive breast cancer and 15 cases of benign lesions; 11 cases of fresh normal breast,10 cases of fresh benign lesions and 10 cases of fresh invasive breast cancer were collected; The expression of B7-H1, PD-1 and IL-10 was measured by immunohistochemistry SP methods in 11 cases of normal breast, 25 cases of benign lesions and 277 cases of invasive breast cancer; The expression of B7-H1, PD-1 and IL-10 was measured by Westerm Blotting in 11 cases of fresh normal breast, 10 cases of fresh benign lesions and 10 cases of fresh invasive breast cancer; Analyze the relationship between the expression of B7-H1 , PD-1 and IL-10 and the pathological characteristics.Results:1. The positive rate of the expression of B7-H1 in breast cancer (79.06%) is higher than that in normal breast (36.36%) and benign lesions(44.00%), the difference has statistical significance(P=0.003 and P<0.0001). There is no statistical significance on positive rate of the expression of B7-H1 between normal breast and benign lesions(P>0.05).2. The positive rate of the expression of PD-1 in normal breast (36.36%)is lower than that in benign lesions (84.00%)and breast cancer (80.51%), the difference has statistical significance(P=0.008 and P=0.002).There was no statistical significance on positive rate of the expression of PD-1 between benign lesions and breast cancer (P>0.05).3. The positive rate of the expression of IL-10 in normal breast (54.55%)is lower than that in benign lesions (88.00%)and breast cancer (92.78%), the difference has statistical significance(P=0.04 and P<0.0001).There is no statistical significance on positive rate of the expression of IL-10 between benign lesions and breast cancer (P>0.05).4. There was no statistical significance between the positive rate of the expression of B7-H1 and age,tumour size,histological classification(P>0.05). The positive rate of the expression of B7-H1 in histological gradeⅠis lower than that in histological gradeⅡandⅢ, the difference has statistical significance(P=0.011 and P=0.002). There is no statistical significance on positive rate of the expression of B7-H1 between histological gradeⅡand histological gradeⅢ(P>0.05). The positive rate of the expression of B7-H1 in the lymph node matastasis group is higher than that in the no lymph node matastasis group , the difference has statistical significance(P<0.0001).5. There is no statistical significance between The positive rate of the expression of PD-1 and age,tumour size,histological classification(P>0.05). The positive rate of the expression of PD-1 in histological gradeⅠis lower than that in histological gradeⅢ, the difference has statistical significance(P=0.039). There is no statistical significance on positive rate of the expression of PD-1 between histological gradeⅠand histological gradeⅡ, as well as histological gradeⅡand histological gradeⅢ(P>0.05). The positive rate of the expression of PD-1 in the lymph node matastasis group is higher than that in the no lymph node matastasis group, the difference has statistical significance(P<0.0001).6. There is no statistical significance between The positive rate of the expression of IL-10 and age,histological classification,histological grade,lymph node matastasis (P>0.05). the positive rate of the expression of IL-10 in group of diameter≥2.5cm is higher than that in the group of diameter<2.5cm, the difference has statistical significance(P<0.0001).7. B7-H1 was positive correlation with PD-1(r=0.590,P<0.0001).IL-10 is not correlated with B7-H1(r=0.004,P=0.953)and PD-1(r=0.004,P=0.953).8. The levels of B7-H1 protein were 0.89±0.12 in normal breast, 0.79±0.23 in benign lesions and 1.07±0.12 in breast cancer respectively. There was no statistical significance on the levels of B7-H1 protein between benign lesions and normal breast(P>0.05).The levels of B7-H1 protein in breast cancer was signifienatly higher than that in normal breast(P=0.010) and benign lesions(P=0.013). The levels of B7-H1 protein in breast cancer was not assotiated with age,tumour size,histological classification,histological grade,Lymph node matastasis(P>0.05).9. The levels of PD-1 protein were 0.89±0.18 in normal breast, 0.96±0.15 in benign lesions and 1.07±0.11 in breast cancer respectively. There was no statistical significance on the levels of PD-1 protein between benign lesions and normal breast,as well as benign lesions and breast cancer(P>0.05).The levels of PD-1 protein in normal breast was signifienatly lower than that in breast cancer (P=0.008). The levels of PD-1 protein in breast cancer was not assotiated with age, tumour size, histological classification, histological grade, Lymph node matastasis(P>0.05).10. The levels of IL-10 protein were 0.84±0.20 in normal breast, 1.01±0.23 in benign lesions and 1.13±0.05 in breast cancer respectively. There was no statistical significance on the levels of IL-10 protein between benign lesions and normal breast, as well as benign lesions and breast cancer(P>0.05).The levels of IL-10 protein in normal breast was signifienatly lower than that in breast cancer (P=0.003). The levels of IL-10 protein in breast cancer was not assotiated with age, tumour size, histological classification, histological grade, Lymph node matastasis(P>0.05).Conclusions:1. The expression of B7-H1,PD-1 and IL-10 in breast cancer is higher than that in normal breast and in benign lesions.2. There is no statistical significance between The positive rate of the expression of B7-H1,PD-1 and age, tumour size, histological classification, however, there is statistical significance between the positive rate of the expression of B7-H1 and histological grade, Lymph node matastasis. There is no statistical significance between the positive rate of the expression of IL-10 and age, histological classification, histological grade, Lymph node matastasis, however, there is statistical significance between The positive rate of the expression of IL-10 and tumour size. B7-H1 , PD-1 and IL-10 was conduced to the judgement of breast cancer.3. B7-H1 is positive correlation with PD-1.IL-10 is not correlated with B7-H1 and PD-1.
