Background:Oxidant/antioxidant imbalance play an important role in the pathogenesis of pulmonary fibrosis.γ-glutamyl cysteine synthetase (γ-glutamylcysteine synthetase,γ- GCS) is the rate-limiting enzyme of glutathione (glutathione,GSH) synthesis.It was considered as very important antioxidant enzyme.Studies in vitro suggested that nuclear factor erythroid-derived-associated factor -2 (NF-E2 related factor 2,Nrf2) controll the expression of anti-oxidase.Studies sugessted Matrine has the effect of anti-fibrosis.And matrine has the effect of anti-fibrosis.Objective: By investigated the chang of the expression of Nrf2 andγ-GCS in the Bleomycin-Induced Pulmonary Fibrosis rats after treated with Matrine.Study the effect and perhaps mechanism in the treatment of matrine in pulmonary fibrosis.Methods:120 adult male SD rats were randomly divided into control group (group A),Bleomycin group (group B),Prednisone Acetate group (group C),Matrine50mg group(group D) and Matrine100mg group(group E).Pulmonary fibrosis model was established by injected with Bleomycin by intratracheal.Detected the concentration of 8-iso-prostaglandin in the plasma.The expression of Nrf2 andγ-GCS mRNA in lung tissue were measured by reverse transcription-polymerase chain reaction(RT-PCR).The protein expressions of Nrf2 andγ-GCS were measured by Western blot.Results:(1)The concentration of 8-iso-prostaglandin was significantly higher in the else groups than group A at 7d and. 14d.And it was the highest in the group B(P<0.01).At 28d,it was the highest in the group B(P<0.01),and the lowest in the group A and group E.There was unsignificant differences between group A and group B(P>0.1).It reached the peak at 14th (P<0.01).(2) Contrast to group A,The protein expression ofγ-GCS and nrf2 were significantly higher in the lung tissue in the else groups.Group B was lower than Group C,D and E(P<0.01).It reached the peak at 7st (P<0.01).(3) Contrast to group A ,The expression ofγ-GCS and nrf2mRNA were significantly higher in the lung tissue in the else groups.Group B was lower than Group C,D and E(P<0.01).And they reached the peak at 7st (P<0.01).(4)They were positively correlated between the expression ofγ-GCS protein and the expression of Nrf2 protein,mRNA.Conclusions:1. Matrine inhibit pulmonary fibrosis in rats induced by bleomycin.2. By controlling the expression of Nrf2 to indirectly control the expression ofγ-GCS. This is a possible mechanism why matrine inhibit pulmonary fibrosis in rats.
