Objective We observe the express variation of zinc finger transcription adjection protein KLF4(Kruppel-like factor 4) in human laryngeal squamous cell carcinoma tissue and polyp of vocal cord tissue, preliminary analyse the effect of KLF4 in pathogenesy of human laryngeal squamous cell carcinoma, to provide the experiment evidence for pathogenesy of human laryngeal squamous cell carcinoma.Methods 1,we used immunohistochemistry stain to deal with 45 laryngeal squamous cell carcinoma tissues (glottic type)which had final diagnosis by pathology, to observe the distribution and expression of KLF4,use 48 polyp of vocal cord tissues as the comparison(control group). According to the ratio and stain intensity of positive cells, we make semiquantitative integral calculus. 2,We use Western-bloting to detect the content of KLF4 in 12 laryngeal squamous cell carcinoma tissues(glottic type) which had final diagnosis by pathology, and 12 polyp of vocal cord tissues.Use theβ-actin as inner comparison. We detect experiment result gray scale value by Image-pro plus Version5.0.1 picture analyse system. All results had been analysed by SPSS13.0 statistic analyse software, P<0.05 have statistical significance.Results 1,immunohistochemistry stain: the positive express ratio is48.9%(31/45) in laryngeal squamous cell carcinoma. The positive express ratio in polyp of vocal cord is 89.6%(43/48). The difference between them have statistical significance, p <0.05. 2,Western bloting:we use the absorbance value of Image-pro plus Version5.0.1 picture analyse system ,find that the expression of KLF4 in laryngeal squamous cell carcinoma tissues is lower than in polyp of vocal cord tissues, p <0.05, the express difference between them have statistical significance.Conclusion 1,The express of zinc finger transcription adjection protein KLF4 in epithelia of polyp of vocal cord tissues is widely, it is primary distribute in cytoplasm and membrane of nucleolus, and it is stably expressing in the polyp of vocal cord tissues .2,The expressing of protein of KLF4 in human laryngeal squamous cell carcinoma tissue(glottic type)is lower than in polyp of vocal cord tissues, the difference between them have statistical significance.
