Objective: To obtain a MGC-803 cell line expressing FHIT protein by transfection ,and investigate the influence of exogenous FHIT gene expression on gastric cancer cells MGC-803 apoptosis induced by octreotide and the molecule mechanism.Methods: Human gastric cancer cells line MGC-803, in which there is expression loss of FHIT gene, was transfected with plasmid pRcCMV-FHIT using lipofectamine transfecting technique. After transfection, Western blot were used to analyse the expression of FHIT genes in positive cells screened out by G418. Each group cells was treated with octreotide at different concentrations. Cell proliferation assay were evaluated by MTT and apoptosis by flow cytometry, as well as the expression of Bcl-2 and Bax was detected with Western blot.Results: (1) Stable expression of FHIT protein in MGC-803 cells was established successful and the expression was detected in MGC-803 cells after transfection with FHIT gene.(2) There was concentration-dependent and time-dependent of octreotide on growth inhibition rate .(3)Results with octreotide treatment 48h later, the apoptotic rates of MGC-803 cells in FHIT gene group (26.777%±1.702%) was significantly higher than that in empty vector group (13.8%±0.511%) and untransfected group (12.634%±0.479%) (F =245.789,P<0.05).(4) The expression of Bcl-2 protein was denclined while the expression of Bax protein was increased in FHIT gene group cells treaed with octreotide.Conclusions: The expression of FHIT gene and octreotide have strong synergistic effect on apoptosis of MGC-803 cell lines,and their action mechanism may relate to the expression of apoptosis-related protein of Bcl-2 family.
