Fragile Histidine Triad Gene Expression In Breast Carcinoma And Its Correlation With P27～(kip1) Protein

Objective: â’ˆTo investigate the effect of p27^kip1 protein and Fragile histidine triad(FHIT) expression on the breast carcinoma. 2. To evaluate the utility of the Fragile histidine triad(FHIT) protein in breast carcinoma in clinic. 3.To study the value of p27^kip1 and FHIT statue for prognosis in breast carcinoma. Methods: By S-P immunohistochemical staining the expressions of Fragile histidine triad (FHIT) gene at protein level and p27 kip1 protein in paraffin-embedded tissues in 40 resected breast carcinoma who are all female from 36 to 60 years old and 6 breast Non-carcinoma who are all female from 24 to 33 years old. There are thirty-one patients whose axillary lymph nodes are positive. Results : In 40 cases of breast carcinoma the positive expressions of p27 kip1 were 18 cases (45%). That of Fragile histidine triad (FHIT) were 15 cases (37.5%). The incidence of positive Fragile histidine triad (FHIT) was 44.4% in the positive p27 kip1 group, While that was 31.8% in the negative p27 kip1 group.(p>0.05) In the invasive duct breast carcinoma the expressions of p27 kip1 protein was 43.6% and that of was 35.9%. In the other classification has too less to be statisticed . In the 31 cases of premenopausal females, p27 kip1(+) was 14 cases (45.5%). Fragile histidine triad(FHIT)(+) was 11 cases (35.5%)(p>0.05). In the 9 cases of postmenopausal females, p27 kip1(+)was 4 cases (44.4%). Fragile histidine triad (FHIT)(+) was 6 cases (66.7%)(p<0.05). The positive rate of p27 kip1 in the patients who have transferation in axillary lymph node was 38.7% (12/31), That of Fragile histidine triad(FHIT) was 29.8%(9/31). (p>0.05) In the positive rate of p27 kip1 in the patients who have Non-transferation in axillary lymph node was 66.7%(6/9),That of Fragile histidine triad (FHIT)(+) was 66.7%(6/9) . (p>0.05) In the 6 cases of distant metastasis of breast cancer , p27 (+) was 1 case (16.7%), Fragile histidine triad (FHIT)(+) was None. In the 34 cases non-distant metastasis of breast cancer , p27^kip1 (+) was 17 case (50%), Fragile histidine triad (FHIT)(+) was 15 case (44.1%) . The incidence of positive p27^kip1 was 60.9%(14/23) in the positive ER group (p<0.05), that of positive Fragile histidine triad (FHIT)(+) was 47.8%(11/23) (p>0.05) . Conclusions: 1.
