Cloning And Sequence Analysis Of The Gastric Carcinoma Related Gene Of Elcox3

Gastric carcinoma is a common malignant tumor, But the exact pathogenesis of gastric carcinoma is not clear so far.The molecular cloning of gastric carcinoma- associated genes and their functional sudies have important meanings.Studies suggested that tumor had high frequency LOH in chromosome 1p35-36,which had many new tumor- associated genes. Igarashi discovered gastric carcinoma had high frequency LOH in chromosome 1p35-36,which had mportant meanings in the development of gastric cancer.In this study ,we successfully cloned the MDSCBC11 gene and also made preliminary studies on their functions by bioinformatical metheds.Methods1 Applying the methods of transcriptional 5'RACE and 3'RACE to determining the transcription start site and the length of the MDSCBC11 represented gene ;2 Using Northern-bot analyze the transcript and size of MDSCBC11 represented gene;3 preliminary studies on their functions analyzed by bioinformatical metheds.Results1 The base of most of 90bp product after the 5'RACE adaptor is A.So the transcriptional start site is A and labelling the A with +1.this transcriptional start site located before the translation site 8bp;2 The length of MDSCBC11 represented gene cDNA is 822bp by the method of 5'RACE and 3'RACE;3 Northern blot analysis revealed that the MDSCBC11 represented gene is expressed as a 800bp transcript in brain,liver,lung,kidney;4 The length of MDSCBC11 represented gene cDNA with an ORF from 8bp to 793bp. The MDSCBC11 represented gene encodes a 29.97 kilo-Dalton protein cmprosed of 261 amino acids.ELCOX3 maps in mitochondria. The deduced amino acid showed 99% homology with COX3,so wo named the gene ELCOX3 .Bioinformatics analysis indicated that ELCOX3 is an acidity protein with a theoretic iso electric point of 6.78;Conclusion1 The transcriptional start site of ELCOX3 gene is A and labelling the A with +1. This transcriptional start site located before the translation site 8bp;2 The length of ELCOX3 cDNA is 822bp and is homologous with COX3 gene.3 The ELCOX3 gene is hingh expressed in normal tissues.there are two transcripts in liver and kidney.
