Objective: To study the expression of hMLH1,RASSF1A,FGF3,Podoplanin and its relationship with clinical Pathology significance on esophageal squamous cell carcinoma. Methods: The expressions of hMLH1 were detected by In situ hybridization in esophageal carcinoma and normal tissues. Immunohistochemical method was used to detect the expressions of RASSF1A,FGF3 and Podoplanin in esophageal carcinoma and normal tissues, to analyse the relationship between them and different clinical pathological factors. Their relationships with prognosis of patients were evaluated by Cox regression model and Kaplan-Meier survival curve analysis. Results: (1) The positive frequency of hMLH1,RASSF1A and FGF3 in esophageal carcinoma tissues is separately 41.67%, 80% and 91.67%,the positive frequency of them in normal tissues is separately 90%, 100% and 50%. The differences have statistically significant (P<0.05). The differences of LVD between esophageal carcinoma and normal tissues have statistically significant. (2) In different clinical stages,different depth of invasion,with or without lymph node metastasis and different degree of differentiation of esophageal carcinoma the positive frequency of hMLH1mRNA have statistically significant (P<0.05). (3) In different clinical stages,different depth of invasion,with or without lymph node metastasis and different length of lesions of esophageal carcinoma the positive frequency of RASSF1A protein have statistically significant (P<0.05). (4) In different clinical stages,different degree of differentiation,different depth of invasion,with or without lymph node metastasis of esophageal carcinoma the positive frequency of FGF3 protein have statistically significant (P<0.05). (5) The differences of LVD in different clinical stages,different depth of invasion,with or without lymph node metastasis and different length of lesions of esophageal carcinoma have statistically significant (P<0.05). (6) The expression of RASSF1A is positively correlated with hMLH1 (r=0.338, P<0.05). (7) The Five-year overall survival rate of the patients is 35%. Univariate survival analysis suggestes that the five-year survival rates between the different groups of LVD have statistically significant. (8) Cox multivariate analysis indicated that the invasion depth of esophageal carcinoma,TNM staging and lymph vessel density are the independent prognostic factors affecting survival rate of patients. Conclusion: (1) It is suggested that hMLH1 play an important role in pathogenesis. It is suggested that RASSF1A is a marker of tumor maligness, it to restrain the development of esophageal carcinoma. The expression of FGF3 has a closed relationship with tumor infiltration. (2) LVD with TNM staging and the invasion depth are the independent prognostic factors affecting survival rate of patients. (3) It is possibly that the gene mutation of hMLH1 in coordination with the gene inactivation of RASSF1A in the pathogenesis of esophageal squamous cell carcinoma.
