Expression And Significance Of HMLH1,hMSH2,EphA2 MRNA In Esophageal Squamous Cell Carcinoma In Uygur And Han Patients In Xinjiang

Objective:To investigate hMLH1,hMSH2 and EphA2 mRNA expression and explore the significance of them in the carcinogenesis and metastasis esophageal carcinoma and their relationship with clinicopathological parameters in esophageal squamous cell carcinoma(ESCC)in Chinese Uygur and Han patients in Xinjiang, Methods:The expression of hMLH1,hMSH2 and EphA2 mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR) in esophageal carcinoma and distant non-carcinoma esophageal tissues from 30 Chinese Uygur patients and 30 Chinese Han patients. Results: (1) In 60 cases of esophageal carcinoma and distant non-carcinoma tissues, the positive rate of hMLH1 and hMSH2 expression in esophageal carcinoma tissues were 65.0%(39/60) and 83.3%(50/60) respectively, in distant non-carcinomat tissues was 88.3%(53/60) and 86.7%(52/60), there were significant difference of hMLH1 (χ2=9.130,P=0.003), but there was not in hMSH2(χ2=0.261,P=0.609), the differential expression rate of hMLH1 and hMSH2 between esophageal carcinoma and non-carcinomatous tissues were75.0%(45/60) and 81.7%(49/60) respectively; The expression level of hMLH1 and hMSH2 mRNA were significantly lower in esophageal carcinoma than in non-carcinoma tissues(0.762±0.036 and 1.025±0.047 vs 0.674±0.039 and 0.797±0.035; t =4.438,P =0.000 vs t =2.317,P=0.022); There was not significant difference in the positive rate of hMLH1 and hMSH2 expression between Han and Uygur patients in carcinoma tissues (χ2=3.590,P=0.058 vsχ2=0.480,P=0.488); The expression of hMLH1 and hMSH2 were correlated with tumor staging(both P<0.05), but there were not relationship with depth of carcinoma invasion , lymph node metastasis and differentiation(P > 0.05). (2) In 60 cases of esophageal carcinoma and distant non-carcinoma esophageal tissues, the positive rate of EphA2 expression was significantly higher in esophageal carcinoma than in distant non-carcinoma tissues [71.7%(43/60) vs 46.7%(28/60);χ2=7.761,P=0.005],the differential expression rate of EphA2 between carcinoma and non-carcinoma tissues was 56.7%(34/60); The expression level of EphA2mRNA was significantly higher in carcinoma than in non-carcinoma tissues(0.269±0.0401 vs 0.153±0.023; t=2.524,P=0.013); There was significant difference in the positive rate of EphA2 expression between Han and Uygur patients in carcinoma tissues [56.7% (17/30) vs 86.7%(26/30);χ2=6.648,P=0.01); The expression of EphA2 mRNA was correlated with carcinoma the depth of invasion and lymph node metastasis(both P<0.05), but not correlated with differentiation levels and tumor staging(both P>0.05). (3) The expression of EphA2 was negatively correlated with hMLH1 and hMSH2 expression. Conclusion: (1)The expression of hMLH1 and hMSH2 was low in esophageal carcinoma tissues , their expression was higher in early stage than in intermediate and advanced. It was indicated that that the mutation of hMLH1 and hMSH2 were involved in the carcinogenesis of esophageal carcinoma in early stage. (2)The expression of EphA2 was high in esophageal carcinoma tissues, it was correlated with the depth of invasion,lymph node metastasis and patients'nationality. The over expression of EphA2 prognosticate local infiltration and more easy to invade and metastasize.(3) The expression of EphA2 was negatively correlated with hMLH1 and hMSH2 expression. (4) hMLH1, hMSH2 and EphA2 play important roles in the carcinogenesis and metastasis of esophageal carcinoma .Combined detection of hMLH1, hMSH2 and EphA2 expression may be used to screen for esophageal carcinoma in high-risk population.
