Objective To assess if the serum Complement C3(C3) is related to the metabolic syndrome(MS) and insulin resistance(IR), and which one of C3,ApoB/ApoA1 ratio,hs-CRP is the most closely correlated with MS.Methods 700 adults were enrolled who accepted physical examination in The First affiliated hospital of Chongqing Medical University, aged above 20 years. Weight, height, blood pressure, waist circumferences, fasting plasma glucose (FPG), fasting serum insulin (FINS) and total cholesterol(TC), triglyceride(TG), high-density lipoprotein cholesterol (HDL-c), low-density lipoprotein cholesterol (LDL-c), Apolipoprotein A1, Apolipoprotein B were measured. Body mass index (BMI),ApoB/ApoA1 ratio and HOMA index (HOMA-IR) were calculated. The metabolic syndrome (MS) was defined according to the criteria made by the International Diabetes Federation in 2005.Resultsâ‘ The C3, ApoB/ApoA1 ratio, hs-CRP increased significantly in subjects with single component of MS, MS or insulin resistance(IR)(P<0.001);and C3, ApoB/ApoA1 ratio, hs-CRP value increased significantly as the number of MS components increased (P<0.001).â‘¡Pearson correlation analysis showed that C3, ApoB/ApoA1 ratio, hs-CRP, waist, SBP, DBP, FPG, TG, HDL-c were correlated each other, partial correlation analysis showed that after adjustment for age and gender, C3 was still closely related to every component of MS above after adjusting ApoB/ApoA1 ratio, hs-CRP;however,ApoB/ApoA1 ratio was only related to three components of MS after adjusting C3, hs-CRP; and hs-CRP was only related to two marker of MS.â‘¢Binary logistic regression analysis showed that when one marker of C3, ApoB/ApoA1 ratio, hs-CRP was analysed, age, gender and the other two marker were adjusted, results indicated that compared with low C3 group, the high C3 group was more likely to get MS (OR=3.17, P<0.001), the subject with high ApoB/ApoA1 ratio and high hs-CRP is more likely to get MS than the one with low ApoB/ApoA1 ratio(OR=2.37,P<0.001) and low hs-CRP(OR=1.74,P<0.05) respectively, but it was not found which one of C3,ApoB/ApoA1 ratio,hs-CRP was the most closely correlated marker with MS.Conclusion C3, ApoB/ApoA1 ratio and hs-CRP were all related to MS and its components. Increase of C3, ApoB/ApoA1 ratio and hs-CRP were all valuable markers of MS, but it is not found which one was the most closely correlated with MS. Objective Establish accurate, high sensitive and stable method to test the concentration of reduced form of nicotinamide-adenine dinucleotid (NADH) in human red blood cells, and use this method in clinical research to know the NADH level in red blood cells of type 2 diabetes. Methods Enzyme Cycling Fluorimetry was applied to determine the concentration of reduced form of nicotinamide-adenine dinucleotid (NADH) in human red blood cells using multifunction MPP detection system. After circular reaction of the substrates and enzmys, the concentration of NADH arrived at the tested region.Result The calibration graph for the determination of NADH is in the range of 0.00-0.80pmol,with a linear equation of y = 99.377x+13.481 obtained, r>0.95. The average recovery is 95.5-100.0%, intra assay RSD is 1.00-2.70% and inter assay RSD is 1.89-12.57%. The method is accurate, sensitive and steady. Conclusion The method is accurate, sensitive, steady, and can be used to determine the concentration of NADH...
