Objective:To investigate the effect of jiejufushenning on the expressions of connective tissue growth factor, platelet-derived growth factor-B in rat glomerular mesangial cells cultivated under high glucose.Methods:Rat glomerular mesangial cells(GMCs) were cultured in vitro with cell culture technology, and the two drug-contained serum of model rats were prepared with serum pharmacology method. Then GMCs were cultured with high concentration glucose(HG) and HG plus irbesartan, different dose(high, middle, low) of jiejufushenning for forty-eight hours, and set up normal control simultaneously. The expressions of mRNA, protein of connective tissue growth factor(CTGF), platelet-derived growth factor-B(PDGF-B) were detected by RT-PCR,Western Blot,respectively. Results:Compared with normal control group, the expressions of mRNA, protein of CTGF and PDGF-B were increased significantly after HG stimulated forty-eight hours(P<0.001). Compared with HG group, HG plus irbesartan, and high, middle, low dose of jiejufushenning inhibited the expressions of mRNA, protein of CTGF and PDGF-B significantly after they worked forty-eight hours(P<0.005).Compared with irbesartan group, the inhibition of high, middle, low dose of jiejufushenning groups were more significant(P<0.05), and a certain dose-dependent was shown (P<0.05).Conclusion:GMCs can increase expressions of mRNA, protein of CTGF and PDGF-B induced by high glucose, jiejufushenning can down-regulate the expressions of CTGF and PDGF-B, thus it may benefit to delay the progression of glomerularsclerosis of diabetic nephropathy.
