Part one Pathological changes and effect observation on Implanted Rabbit Breast Tumor Treated with High Intensity Focused UltrasoundObjective: To observe the histopathological changes and prostecdtive efficacy of implanted rabbit breast tumor with high intensity focused ultrasound (HIFU)ablation.Methods: All 62 rabbits were divided into A group (pathology observation group,n=42),they were subdivided into HIFU group (n=35) and control group (n=7) ; B group(effect observation group,n=20), they were subdivided into HIFU group (n=10) and control group (n=10), HIFU ablation was performed for the HIFU group's tumor with ultrasound imaging guidance.After ablation,collect specimens from A group for histopathological examination immediately and on 1st, 3rd, 7th, 14th, 21st, 28th day;The average survival time, survival rate after 10 months and nodes metastasis time of the B group were observed.Results: After HIFU ablation, Immediately the whole tumor tissue appeared gray, the morphology and arrangements of the nucleus of the ablated tumor cells in the HIFU group showed no obvious alteration in HE stained sections, however, electron microscopy revealed that the tumor cells in an irreversible state and nuclear fragmentation, cell organ destroyed and inequality of size of vacuolus appeared in cytoplasm,showed the cells dead,with the prolonging of time,the necrosis of tumor tissue got liquefied and absorbed,replaced by fibrous tissue after 28 days; The average survival time and nodes metastasis time were 214.1±111.6 days,53.3±7.5 days in the HIFU group ,respectively,they were longer than those of control group, the survival rate after 10 months(60.0%) were higher than those of control group, P<0.05.Conclusion: HIFU can effectively destroy the implanted rabbit breast tumor tissue, inhibit tumor's metastasis and prolong the survival span of the experimental rabbit, it provided referred value for clinical application of HIFU in the conservation breast treatment of breast cancer.Part two Effect of expression of tomor associated antigens in breast cancer with High Intensity Focused Ultrasound ablationObjective: To observe the expression of tomor associated Antigens in breast cancer with high intensity focused ultrasound(HIFU) ablation.Methods:21 breast cancer specimens were be diagnosed by pathologic examination,after HIFU ablation, All specimens were be examed by using HE staining and enzyme histochemical staining and immunohistochemical technology immediately.Results:In the target area of HIFU ablation, histopathologic observation showed vacuole-like structure in cytoplasm, and there were no significant changes in cell's contour and nucleus karyotype; The expression of Estrogen receptor(ER), Progesterone receptor(PR), cerbB-2 and P53 were positive in the normal cancer tissues,but they were negative in the target area of ablated cancer tissues, then only found that expression of ER in two specimens and PR in one specimen were weakly positive at junctional zone between the ablated target area and not ablated one,ER,PR,cerbB-2 and P53 were negative in the other specimens;succinate dehy drogenase(SDH) was positive in not ablated area of cancer tissues, so was negative in the target area and junctional zone of all specimens,showed the cells dead.Conclusion: HIFU ablation can destroy the breast cancer cells,and expression of tomor associated antigens are decreased or negative,Some tumor antigens remained after HIFU ablation,this could provide a potential antigen source to stimulate antitumor immune response and generate the unique cellular immune responses against tumor cells.
