Purpose To investate the effect of ORI on the gene expression profile of human esophageal cancer cell line SHEEC by gene chip.Method SHEEC cells were treated with 32μg/ml ORI for 1h and 8h respectively as experimental group, SHEEC cells were not treated with ORI as control group. The effect of ORI on the gene expression profile of human esophageal cancer SHEEC cells were investated by gene chip. In gene expression data, the differentially expressed genes in SHEEC cells treated with ORI for 1h and 8h were screened according to signal log ratio (SLR)≥1 and≤-1. The differentially expressed genes were classified according to gene ontology biological process. Screening the differentially expressed genes according to the following criterias:①. 20 of the greatest up-regulated genes of SHEEC treated with ORI for 1h or 8h,and 20 of the greatest down-regulated genes of SHEEC treated with ORI for 1h or 8h. Repeating to detected genes expression by gene chip,the high SLR of the greatest up-regulated genes described above were greater than 1,and so were the low SLR; the high SLR of the greatest down-regulated genes described above were less than -1,and so were the low SLR. The differentilly expressed genes in SHEEC cells treated with ORI for 1h and 8h meanwhile. Repeating to detected genes expression by gene chip, the high SLR of the up-regulated genes described above were greater than 0.7,and so were the low SLR; the high SLR of the down-regulated genes were less than -0.7,and so were the low SLR.②. annotation of Genes GO were completed.③. Biological process of the differentially expressed genes are related to apoptosis, signal transduction and transcription regulation. And the genes are detected by fluorescent quantitation RT-PCR.Result SHEEC cells were treated with 32μg/ml ORI for 1h. A total of 1286 genes were changed, 346 of which were up-regulated, and 940 genes down-regulate;SHEEC cells were treated with 32μg/ml ORI for 8h. A total of 3856 genes were changed, 1519 of which were up-regulated, and 2337 genes down-regulated. A total of 280 genes were detected with a difference of expression levels more than 2 time at 1h, 23 of which were up-regulated and 257 down-regulated. A total of 737 genes were detected with a difference of expression levels more than 2 time at 8h, 166 of which were up-regulated, and the down-regulated. According to gene ontology biological process, the differentially expressed genes were related to cell signal transduction, transcriptional regulation, apoptosis, metabolism, material transport, cell adhension, cell proliferction, defense response, RNA processing and so on. Fluorescent quantitative RT-PCR had confirmed 22 of differentially expressed genes which were analysised and screened by the criterias. There were 12 genes with statistical significance.Conclusion There are many genes involved in ORI inducing SHEEC cells apoptosis through mitochondrial pathway, which are mainly related to apoptosis, signal transduction and transcriptional regulation.
