Objective : Brain concussion is a transient disturbance of brain function immediately after an outside force on head, which can recover automatically. The typical symptom of brain concussion is a transient conscious loss and a short-term memory disturbance, having no significant changes in pathomorphology and no obvious physical sign of injury in nervous system. So the diagnosis and therapy is very difficult. In general, the clinic have three points to diagnose this disease, which is outside force, conscious loss and memory disturbance . But many investigators suggest that the brain concussion is caused by slight diffuse axonal injury (DAI), furthermore it also have some changes in pathomorphology. In this study, the rat was strike by artificial hammer to make an animal model of brain concussion to investigate the pathogenesis of brain concussion, the oxidative stress and the expression of inflammation factor in brain tissue. Furthermore the serum biochemistry index was measured. In order to search the medicine of relieving brain injury ,we infused CCK-8 to the rat before strike and investigate the alteration of above index.Methods: The animal model of brain concussion was established by the artifical hammer falling in 135° and striking the external occipital protuperance of the rat to make a immediate conscious loss of 1-3 min , those rats quickly recovered without any cerebral hemorrhage in neuropathy .Ninety healthy wistar rats of either sex, weighting 200g were randomly divided into three groups: control group , brain concussion group, and CCK-8 group . During different time after strike the level of MDA ,SOD , CRP was measured and the expression of TNF-α,IL-1β in brain tissue was examined by immunochemistry, meanwhile the histopathological changes of brain tissue was observed. Then we infused CCK-8 to the rat before strike and examined the above items .Results:1 Change of pathological section There was no alteration in brain tissue in control group.In brain concussion group slight congestion and cerebral edema were observed in pathological section. Nerve myelin sheath became disorder,disruption, gryposis and swelling in callus,hippocampi and brain stem at 6h. at 12h and 24h the above changes were more severe .In CCK group there were no significant alteration compared with that in strike group. 2 Changes of MDA contents and SOD activity in brain The content of brain MDA in control group was1.10±0.11 nmol/mg prot, in brain concussion 2h group was 2.27 ± 0.94 nmol/mg prot(P<0.01 vs control group), in CCK-8 2h group was 1.37±0.94 nmol/mgprot(P<0.05 vs brain concussion 2h group).The SOD activity of brain in control group was 13.15±2.07 NU/mg prot, strike 2h group was 11.12±0.79 NU/mg prot(P<0.05 vs control group), CCK-8 2h group was 12.81±1.27(P<0.05 vs brain concussion 2h group ).3 Immunochemistry in brain tissue.In control group TNF-α,IL-1β did not express or little express in Cerebral cortex and hippocampus . But in brain concussion group the expression of TNF-α and IL-1β in above position increased significantly at 6h and 12h following strike.In CCK group the expression of TNF-α and IL-1β were lower compared with those in brain concussion group, though still increased significantly compared with control group .4 Changes of MDA contents and SOD activity in serum The content of serum MDA in control group was 5.22±0.63 nmol/ml, in brain concussion 2h group was 6.59± 0.86 nmol/ml(P<0.01 vs control group), in CCK 2h group was 5.33±0.86 nmol/ml(P<0.05 vs strike group)。The serum SOD activity in control group was 291.84±58.11(NU/ml),in strike 2h group was 176.00±18.99 NU/ml(P<0.01 vs control group),in brainconcussion 6h group was 205.61 ±103.71 NU/ml(P<0.05 vs control group),in CCK 2h group was 232.16±41.47 NU/ml(P<0.05vs brain concussion 2h group),in CCK 6h group was 256.83±49.06(P>0.05vs strike group). 5 Changes of serum CRP As compared with the control group (1.59±0.54...
