Immunohistochemical Study Of Mast Cell Tryptase And Other Mediators In Some Tissues Of Persons Died Of Anaphylactic Shock

Mast cells (MC) are one of important immuno cells, which can secret many bioactive substance ,such as histamine,5-HT,SRS-A and heparin . They have a pivotal role in allergic reactions and take part in many other pathophysiological conditions such as wound healing, fibrosis, tumors and autoimmune diseasesMC 's surface express highly affinity acceptors of IgE,the cross-linking of which is playing a key role on the activation and degranulation of MC. The binding of IgE and their acceptors through a series of transmission of signals trigged the degranulation of MC .When MC are activated,they can release a n array of secrtions. Tryptase and chymase , important and highly specific proteins in MC, are important markers to differenit of MC subtype,while basophils have little of them .Tryptase is a kind of serine proteases which can be found in many mammal's MC as an active granule .Compared with histamine,tryptase is a more selective marker of MC .Thus ,tryptase are frequenty regarded as a characteristic marker of human MC presently. MC can producemany cell activing factors.Now we have found after the activation of MC , it can synthesize many proinflamation factors, such as TNF-a, IL-1,IL-6,GM-CSF and IFNy. Through the secreation of these cytokines,MC take part in many pathophysiological conditions.Anaphylactic shock,especially drug-induced anaphylactic shock, is one of clinical emergence.lt occurs suddenly,difficult to predict and threat the lives of patients.The drug often induces anaphylactic shock include antibioticantibiotic, serum, antitoxin , local anesthetic and antipyretic analgesic .Drug induced anaphylactic shock is one of important reasons of medical tangle. While the current diagnosis standard mainly according to clinical symptom and objective signs.Because it has no exactly laboratory diagonosis.,some medical tangles are often not solved for a long time. Thus ,it has become a urgent problem of forsensic pathology that studying the mechnism of anaphylactic shock ,finding a exactly diagnosis standard and putting out a easy to use diagnosis method.This experiment study the distribution of MC when anaphylactic shock occurred through the immunohistochemistry of tryptase andIL-1 P and toluidine blue staining of 27 cases died of anaphylactic shock and 8 cases died of other diseases. The results are as follows:1 HEPeople died of other diseases :lung and intestinal mucosa are normalPeople died of anaphylactic shock::lung is congestion and edema, alveolar ectasia and the intervals collaped forming lung edema The alveoli are filled with exudates.Bronchial muscosa is infiltrated by eosinophils. Lamina propria of intestinal mucosa is infiltrated by large number of eosinphils. 2 tryptasePeople died of other diseases :there is no positive cell in lung and intestinal mucosa.People died of anaphylactic shock:there are a lot of positive cells in lung ,mainly around the vessels. There are a lot of positive cells on intestinal mucosa.
