Purpose: Vascular endothelial growth factor (VEGF) expression is thought to play a critical role in mediating physiological and pathological angiogenesis.Furthermore, present study indicated that VEGF modulates skeletal myoblast function and plays a key role in myoblast migration and survival during skeletal muscle repair. The present study investigated the time-course changes in VEGF protein expression and capillarity in rat gastrocnemius muscle as well as exercise-induced muscle damage situation after a single acute exhaustive exercise bout,and attempt to investigate the possible relations between them.Method:Sixty adult male Sprague-Dawley rats are divided into ten groups at random:control group(n=6);Single bout acute treadmill exercise group(n=54),which was further divided into 0h,1h,2h,3h,4h,1day,3day,5day,10day postexercise groups. Exercise group performed 200-min treadmill exercise at 16 m/min,on a -16°incline, with a 5-min rest period between two 100-min running bouts.Result:(1) In rat gastrocnemius muscle, VEGF protein level increased in the Single bout acute treadmill exercise group at 0h postexercise ,reached the peak 2 hours after exercise,and decreased gradually from 3h to 2day,interestingly, VEGF protein level increased again 3 days after running,on 5th and 10th day VEGF protein level return to base level,showed the bimodal expression pattern.(2)The most serious damage of rat gastrocnemius muscle showed on the 1st and 3rd days after one bout of exercise,but the damage became less serious on 10th days and no further damage was seen in the microstructure of gastrocnemius muscle .(3)Skeletal muscle capillaries were observed by light mcroscope,the results showed that capillaries obviously increased on 3rd,5th and 10th day...
