Exercise may improve the heart function effectively. The different modes of exercise can cause organism to have the varying degree stress. FOS protein and HSP70 are two kinds of proteins expressing obviously in the stress situation. The main purpose of this topic research is that comparing and analyzing the different changes and the common rules of FOS protein and HSP70 expression through the different animal exercise model. Then it researches the regulation mechanism of heart gene proteins in acute exercise and in long-term endurance exercise, and discusses the changes of gene protein expression in exercise-induced cardiac hypertrophy which the long-term endurance exercise causes. Thus it provides the theory basis for the exercise promoting the heart health.This research selects 100 male SD rats, and divides into 10 groups randomly. Each group has 10 rats. Acute exercise: control group, 0 hour, 1 hour, 3 hours, 5 hours, 12 hours, 24 hours after exercise groups; Long-term endurance exercise: control group, 0 hour, 24 hours after exercise groups. Acute exercise groups are trained on the treadmill which refers to the Bedford increasing load model. The long-term endurance exercise groups use exercise program which refers to the Bedford increasing load treadmill and long-term endurance treadmill model planed by Chang Yun . The exercise groups are trained 6 days and rest 1 day every week for 10 weeks. There are three stages in the entire training cycle: The speed and time are added in 1th to 4th week. The slope is added in 5th to 7th week. The speed is added in 8th to 10th week. The rats are killed after the last training according to the various time in the experiment. The blood is determined serum CK; the hearts are took out to weight and cut about 1×1×1mm~3 cardiac muscle in the left ventricle , then are fixed, dehydrated, waxed, embedded, and sliced. They are used in the conventional HE dyeing and examines FOS protein and HSP70.Result discoveries: In acute exercise, it has the mild damage on the cardiac muscle structure. Serum CK increases in 0-3 hours after exercise mildly, and ascends in 5 hours after exercise obviously. Serum CK achieves the peak level in 12 hours after exercise, and continues to 24 hours after exercise. FOS protein increases mildly in 0 hour after exercise, and obviously increases in 1 hour after exercise. FOS protein achieves the peak in 3 hours after exercise, and restores to level of control group in 5 hours after exercise. HSP70 mildly increases in 0 hour after exercise, and obviously increases in 1 hour after exercise and 3 hours after exercise. HSP70 achieves the peak in 5 hours after exercise, and continues to 12 hours after exercise, and restores to level of control group in 24 hours after exercise; The long-term endurance exercise causes the cardiac muscle structure to have the positive compatible changes. Serum CK is not appeared the significance change. FOS protein is remarkably increased in 0 hour after exercise. FOS protein is reduced in 24 hours after exercise, but still is higher than the control group. HSP70 obviously increases in 0 hour after exercise group and 24 hours after exercise group.Conclusion: 1. Acute exercise may cause the cardiac muscle structure to damage mildly. The long-term endurance exercise causes the cardiac muscle structure to have the positive compatible change; 2. After acute exercise stress, FOS protein can express fast and early, also the expression time is short. After acute exercise stress, HSP70 also can express fast, but the expression peak of HSP70 is later than that of FOS protein, and the expression time is slightly longer. FOS protein expression induces HSP70 expression; 3. Long-term endurance exercise stress causes FOS protein massively expressions. The activation of FOS protein starts the cardiac structure protein to express. It appears cardiac hypertrophy. It is a starting factor for cardiac hypertrophy; The long-term endurance exercise induces the HSP70 expression highly. It is helpful to reduce the damage of cardiac muscle, and to promote damage restoration.
