| EPIDEMIOLOGYDiabetic gastroparesis (DGP) is one of the most common complications of diabetes. According to statistics,75% of diabetics have DGP. The pathogenesis of DGP associated with RAGE, interstitial cells of cajal, autonomic neuropathy, enteric nervous system, blood glucose, and smooth muscle and so on. The above factors closely affect the development of DGP.Tong xiaolin create Tangweianfang from xiaobanxiatang and zhizhuwan, to control blood sugar and alleciate gastrointestinal symptoms, protect the gastrointestinal motility. This essay discusses its possible mechanisms to increase scientific basis.METHODS8 of 40 wistar male rats were randomly assigned to the normal control group, and the remaining was injected with 1% STZ. Diabetic rat model is considered established if the fasting plasma glucose≥16.9 mmol/L three days after injection. Successful modeling rats were divided into four groups according to their weigh and fasting blood glucose:model group, high dose group, middle dose group and low dose group. Recorded weight every week and fasting blood glucose every four weeks. After 8 weeks, RAGE, AGEs were determined by immune histochemistry. RAGE, S100B, c-kit, SCF, PSD95were determined by western-blotting (WB) and PCR.RESULTS1. GSPThe study has found that blood glucose, glycated serum protein content were different from normal group (P<0.05) 8 weeks after STZ modeling; Tangweian has positive effect on glycated serum protein content with improvement (P<0.05) (P<0.01).2. Immunity histochemistry:Compared with the normal group, model group rat stomach AGE, RAGE expression was strengthened; compared with the model group, treatment group with improved diabetic rat stomach AGE, RAGE expression effect.3. PCR:The expression of RAGE, S100B in model group were increased compared with normal mice. Compared with model group, treatment group decreased (p=0). The expression of c-kit, SCF, PSD95 were decreased compared with normal mice. Compared with model group, treatment group increased obviously (p=0).4. Western blot:Compared with the normal group, The expression of quality of c-kit and SCF, PSD95 in model group were decreased compared with normal mice. Compared with model group, treatment group increased.CONCLUSIONTangweian can reduce STZ-induced diabetic rats blood sugar, reduce the content of RAGE, AGEs, S100B, protect interstitial cells of cajal, promote the nerve conduction, and provide a reference for DGP neuromodulation treatment. |
