Effect Of Angelica Sinensis Decoction On Immune Function In Mice With Repeated Respiratory Tract Infection

Objective:Clinical aspects mainly through analysis of clinical cases,summarized tutor treatment of recurrent respiratory tract infections experience--rom blood stasis and heat accumulation,tr eating and using the classic recipe dangguiliuhuang soup treatment;experiments is done main ly by intraperitoneal injection of LPS (lipopolysaccharide) establish heat stasis recurrent resp iratory tract infection within the knot syndrome mouse model,and then uses dangguiliuhuang soup intervention on the model, observation model rat signs changes before and after the experiment, the change of microcirculation and immune factors, in order to evaluate recurre nt respiratory tract infections of blood stasis and heat accumulation model,and reveal the eff ect of dangguiliuhuang soup on the rat model of Thl and Th2 cytokines.Methods:60 ICR mice were randomly divided into normal group, model group, the Chinese me dicine group (dangguiliuhuang soup group) and western medicine group(pidotimod group).Ad opted by intraperitoneal injection of normal saline (NS) in normal group,intraperitoneal injec tion of LPS 2mg/kg (with saline formulated into 2mg/ml) in the other groups to form the model.Normal group and model group were given NS,Chinese traditional medicine group we re given dangguiliuhuang soup group,western medicine group were given pidotimod(10ml/kg/ d),continuous administration of four weeks.Observing body weight,body temperature changes in the tongue,tail petechiae,ear and zhao jia color,sweating and other characterization,and te sting ear and tail microcirculation,and then measured the IL-4,IL-12,IL-10,INF-γ content in mice serum by ELISA method.Results:1.Establishment and evaluation of blood stasis and heat accumulation modelCompared with the normal group, model group showed significant sweating, irritability, dry stool, tongue purple, zhao jia purple and tail significant bruising, body temperature increased(P<0.05),mice ear profile, tail microcirculation and weight decreased significantly(P<0.05), showing the characteristics of blood stasis and heat accumulation.2.The change of dangguiliuhuang soup jiawei on rat model of blood stasis and heat accumulationCompared with the model group, Chinese medicine group decreased temperature(P<0.0 5),increased weight (P<0.05);ear and tail microcirculation increasing(P<0.05).3.Effect of dangguiliuhuang soup jiawei on model rat Th1 and Th2 cytokines1. Compared with the normal group, model group interleukin 4, interleukin-10 levels were reduced, interleukin-12, interferon gamma levels increased, the difference is statistically significant (P<0.05);2.compared with the model group, Chinese medicine group can significantly decreased i nterleukin 4 and interferon gamma levels,improved interleukin 12 and interleukin 10 levels, the difference were statistically significant (P<0.05).Conclusion:1.Intraperitoneal injection of LPS to establish mice model of heat and blood stasis in sticki ng showing IL-4,IL-10 were reduced,IL-12,IFN-γ were elevated;that this process in Th1 cell response is dominant and Th2 type cells were at a disadvantage, cell balancing was destro yed.2.Dangguiliuhuang soup of heat-blood stasis within the node type recurrent respiratory tract infection (RRTI) model mice Th1 and Th2 type cytokines has moderating effect,increased of Th1 type cytokines IL-12 and Th2 cytokine IL-10,lowered the Th1 cytokine IFN-γ and Th2 cytokine IL-4;ensure the pathogen of the immune response,balance the level Th1 and Th2 cells,so as to control the occurrence and development of the disease.
