Ginseng fruit is the ripe fruit of traditional valuable Chinese medicine ginseng(Panax ginseng C.A. Meyer).The polysaccharide is one of the main active ingredient of ginseng fruit, has anti-aging, anti-lipid peroxidation and can improve body’s ability to adapt to the environment and defense capabilities. As the main producing country of ginseng, fruit pulp production of China is rich. But there were few studies on the ginseng fruit polysaccharide, which was limited to the simpale separation and the whole activity research. The systematic and standardized extraction, separation, structure analysis, structure-activity relationship research was lacked. Based on the blank research of ginseng fruit polysaccharide and the rich resources of our country, the separating method for ginseng polysaccharide was innovated. Ginseng fruit Water Soluble Total Polysaccharide was purified Comparehensively and Systematically by online monitoring separation system. And the monosaccharide compaosition, molecular weight, IR of average molecular fraction was determinated. In addition, anti-tumor activity of each second fraction was explorated preliminarily. The main research are as follows:1.Extraction and Purification of Ginseng fruit PolysaccharideFirstly, the Ginseng fruit total polysaccharide WPGF was extracted by water extraction and alcohol precipitation method; Secondly, based on the ultrafiltration-cellulose column chromatography-gel column chromatography separation model, Ginseng fruit Water Soluble Total Polysaccharide was purified Comparehensively and Systematically by online monitoring separation system. Finally obtain one first fraction of Ginseng fruit polysaccharide (WPGFH) by ultrafiltration, four secondary fractions by cellulose column chromatography (WPGFH-0, WPGFH-0.04, WPGFH-0.08, WPGFH-0.12), eight Third fractions by gel column chromatography (WPGFH-0-1, WPGFH-0-2, WPGFH-0.04-2, WPGFH-0.04-3, WPGFH-0.08-1, WPGFH-0.08-2, WPGFH- 0.12-1, WPGFH-0.12-2), Providing a reference for the separation and purification of plant polysaccharide.2.Preliminary analysis of Ginseng polysaccharide the different fractions of physicochemical propertiesUsing PMP-HPLC and HPGPC preliminary analysis the different fractions of ginseng fruit Polysaccharide’s monosaccharide compaosition, molecular weight homogeneity, molecular weight range, determine the molecular weight range of the Ginseng fruit polysaccharide’s three fractions,the two fractions and three fractions of the WPGFH-0.08-1 monosaccharide compaosition, and analysising the WPGFH-0.08-1 with the IR. we concluded that it may be arabinogalactan pectin. Provide the basis for further separation and precise structure of Ginseng Polysaccharide analysis.3.Preliminary inquiry Ginseng Polysaccharide’s Antitumor ActivityCompaarison of the four individuals ginseng fruit two fractions of HepG2 hepatoma cells in vitro inhibitory activity, suggesting that ginseng fruit polysaccharide fractions of different HepG-2 cells for growth inhibition is selective, and the fractions WPGFH-0 and WPGFH-MTT assay 0.12 at 0~3mg/mL did not show a significant anti-tumor activity, but the fraction WPGFH-0.04 and WPGFH-0.8 in the dose reaches the tumors showed strong 3mg/ mL inhibitory activity for the full understanding of ginseng fruit polysaccharide the activity of the foundation.The study, first proposed UF-cellulose column chromatography-gel column chromatography combined classification purification model; secondly, based on preparative liquid phase, on-line monitoring of the formation of separation and purification of polysaccharides plat form and apply ginseng fruit overall grade separation and purification of polysaccharides; finally, using the chemical and chromatographic methods for monosaccharide compaosition analysis and determination of molecular weight range, of the secondary and tertiary fractions, combined with the analysis of IR uniform molecular weight compaonent WPGFH-0.08-1 the structure of a preliminary analysis, a preliminary compaarison of each of the two fractions in vitro anti-tumor activity. The conclusions of this issue will provide a theoretical basis for further research and development of Ginseng Polysaccharide, it will also bring a new reference offer separation and purification plant polysaccharide.
