| Objective1 To investigate the effect of different concentrationDahuangzhechong pilldecoction on cell cycle and proliferation in breast cancer MCF-7 cells and colorectal cance LOV O cells.2 Through analyzing the clinicalliterature about treating tumor with DahuangZhech ongpill, and evaluating the quality of included studies, to explore the application pros pect of ZhangZhongjing’sclassical prescription"DahuangZhechongPill"in the Derma to lo gical Department.Method1 Using optical microscopy, we observed the changes of cellular morphology in MCF-7 cells and colorectal cance LOVO cells treated by different concentrationDahuan gzhechong pill decoction. MTT colorimetric assay was applied to detect cell proliferati on in MCF-7 cells; Flow cytometry was used to analyze cell cycle in MCF-7 cells2 We searched the CNKI(2000-2015), Wanfang database(2000-2015), Collected the clinicalliterature about using DahuangZhechongpill to treat tumor, analyzed the appl ied range of DahuangZhechongPill in the Derma tological Department, as well as the usage, the effect, the follow-up,these curity and soon, then calculated the applicationp rospect, and also evaluated the quality of included studies.Result1 MTT results showed that MCF-7 cells treated byevery concentrationDahuangzhe chong pill decoction had obviouslyinhibiting effect on cell proliferation, the cell inhibit ion rate could be up to 89.2% at 160mg·ml-1, visible under an optical microscope, cel 1 morphology, cell death a lot but with the concentration increased, the inhibitory effe ct gradually decreased, concentration of 1280mg·ml-1, the inhibition dropped to 18.7%. Flow cytometry results showedthe percentage of G1, S and G2 phase cells treated by different concentration Dahuangzhechong pill decoction had significant difference com pared with the control group. The percentage of S phase cells could be increased in LOVO cells treated by 160,640mg·ml-1 and 1280mg·ml-1Dahuangzhechong pill decocti on; However, we found the percentage of G1 phase cells could be decreased and the percentage of S phase cells could be increased.in MCF-7 cells with 160mg·ml-1Dahuan gzhechong pill decoction.2 In the included 32 articles, there was 24 RCT,including 4 high quality studies , the others were low quality studies.20 studies had moderatebias possibility,4 studies had high biaspossibility. Those articles involved 8 kinds of tumor especiallyacne, Hep atic fibrosis of liver cancer, hysteromyoma, hyperplasia of mammary glands, ovarian c yst. Using DahuangZhechongPill combined with other drugs was more extensive, the d osage of DahuangZhechongPill was mainly unified,the course of treating non acute tu mor was often 12 weeks.The totale ffective rate was 42%～100%, the long term thera py was need more further follow-upobservation.Conclusion1 We found Dahuangzhechong Pill decoction of breast cancer MCF-7 cell prolifer ation was inhibited, in a concentration range of 80mg · ml-1-1280mg · ml-1 of Dahu angzhechong pill decoction has obvious inhibit MCF-7 cell proliferation, the inhibition increases as the concentration of waning. Moreover, we also found that S phase arres t could be induced byDahuangzhechong pill decoction. Our findings can provide new knowledge aboutDahuangzhechong pill decoction in clinical therapy of Breast cancer.2 The limite devidence indicates that DahuangZhechongpill has been more and m ore widely used in the treating tumor, it can be used for treating 10 or more kind of tumor, and is often used with other drugs, and it has a good curative effect, little un toward reaction, high safety, and a good Application prospect. But there is few related clinicalstudies, and most method logical qualities are not high,most of them may hav e moderate biaspossibility. The more high quality RCT studies are needed. |
