Preventive Effect Of Dahuangzhechong Pill Combined With Bifendate Dripping Pill On Hepatic Fibrosis Induced By CCL₄in Rats

Objective: To observe the preventive effect of Dahuangzhechong Pill combined withBifendate Pill in liver fibrosis by CCL4in rats.Methods:64male SD rats with170g-230gin weight were randomly divided into six groups:Blank group, modelgroup, Dahuangzhechong pill low dose, high dose group, Dahuangzhechong pilland bifendate pill group,Bifendate Pills group. Except the blank group, the other5groups received intraperitoneal injection of20%CCL4modeling. Beginning from the dateof modeling, the groups were intragastric administration intervention. For6weeks, toassess the general state of the rats, hepatic, spleen index,serum alanineaminotransferase(ALT),aspartate aminotransferase(AST) activity,serum transforminggrowth factorβ1(TGF-β1), matrix metalloproteinase2(MMP-2), Liverhydroxyproline(Hyp) content and to observe the structure of liver cell and the degree ofliver fibrosis by Light microscopy.Result: State of each drug group rats compared withnormal group is a bit poor, but better than the model group, the effectof Dahuangzhechong pill high dose group and the combined group significantly. Comparedwith the model group,Dahuangzhechong pill high dose group and the combined groupcan reduce rat spleen index (p <0.05) The combined group the liver index was lower thanthat in model group(p<0.01), Dahuang Zhechong pills high dose group liver index lower than thatof model group (p<0.05); Compared with the model group, the treatment group decreased serumAST, ALT levels (p <0.01); Dahuang Zhechong pill high dose group can reduced the serumTGF-β1(p<0.05), and the combined group canreduced the serum TGF-β1(p<0.01);Dahuang Zhechong pill high dose group and DDB group elevated MMP-2content (P<0.05), the combined group and DDB group elevated MMP-2content (P<0.05);the combined group decreased the content of Hyp in liver tissue of abnormallyelevated (p<0.01); From HE and Masson staining, Dahuang Zhechong pill group of lowdosage treatment effect was not significant, hepatic fibrosis of Dahuangzhechong pill high dose group, bifendate group and combination group were different degrees of improvement.Fiber interval narrowed, Connective tissue hyperplasia reduced inflammatory infiltrationrelieved, and combine group was better. Conclusion: Dahuangzhechong pill combinedwith Bifendate pill in liver fibrosis induced by CCL4in rats has better prevention, and hasgood clinical application and development prospects.
