Blood transfusion is one of the most common treatments in the clinic. Because of the shortage of blood supply, matching complex and creating cross-infection easily, looking for a safe and effective blood substitutes is a key and hot point in the development of medical research in a century. Blood substitute is solution which not only has the function of carrying oxygen, but also expanding. So it must has the traits such as abundant source, without toxicity and immunegenicity, good mechanical rheological properties, long shelf life and be easy physiological metabolism. Blood substitute can be split into two categories:Hemoglobin based oxygen carriers and Perfluorocarbon compounds.The main component of HBOC is ultrapure hemoglobin. Because of losing of SOD, CAT etc, it is easily to be oxidized by ROS. What is more, reperfusion itself could produce ROS which is harmful to the body. Hemoglobin is easily oxidized to HbFe3+, which further induces the generation of reactive oxygen species. Under the continuous action of hydrogen peroxide, Fe3+ Hb and ROS form the circulation system which not only accelerates the reperfusion injury, but also greatly weaken the ability of carrying oxygen. So the problem cries for solution.Because the content of MetHb is one of the main factors affecting blood substitutes carrying and releasing oxygen properties. This paper is mainly divided into two parts:In the first part, we studied the oxygen releasing regularity of hemoglobin and red blood cells, in order to provide reference for the preparation of effective carrying and releasing oxygen of HBOCs. In the second part, we choosed the modifier for Mal-mPEG and nitrogen oxygen free radicals to explore if the introduction of Mal-mPEG and nitrogen oxygen free radicals have any effect on the generation of MetHb, and whether it can inhibit the oxidation of HBOCs or not.Experimental results show that:By determination of oxygen partial pressure in the oxygen release process of human red blood cell or hemoglobin solution with a concentration of 5 g/dL, we found the law of oxygen release of red blood cells and hemoglobin. In vitro, the content of MetHb of HBOCs with Mal-mPEG and nitrogen oxygen free radical as modifier was changed with time, thus the effect of modifier on the autoxidation of Hb was found.
