Psoriasis is a kind of immune abnormality, hyperplasia, inflammatory skin disease which decided by multi gene, induced by multiple environmental factors. The repeated emergence of multilayer dry scaling on the basis of erythema with varying degrees of itching is the main clinical manifestation. It is often repeated attacks, sustained, resulting in serious psychological burden to patients. The etiology and pathogenesis of psoriasis is not clear, the traditional Chinese medicine treatment has achieved certain results, but the mechanism of the treatment of psoriasis of the effective components and monomer of Chinese medicine have yet to be further studied.Paeoniflorin is extract of Radix Paeoniae Rubra and has anti-inflammatory, immunomodulatory, induction of apoptosis, neuroprotective and antioxidant and tranquilizing effect, its mechanism of action may be through the intervention of the NF-κB、p-Akt and STAT3 signaling pathway leading to downregulation of IL-1β、IL-6. TNF-α、IFN-γ inflammatory factor and induce T cell differentiation and apoptosis.Paeonol is extract of Paeonia suffruticosa and has anti-inflammatory, neural protection, antiproliferative, antioxidant effect, also it protects vascular. The mechanism of paeonol may be intervention of MAPK/ERK and p38 signal transduction pathway and inhibition of MAPKs/NF-κB pathway and disrupt the IL-6 mediated STAT3 path leading to downregulation of TNF-α, IL-1, IL-6, IL-10 cytokine induced apoptosis.The Suppressor of cytokine signaling (SOCS) family of proteins represent one important mechanism to extinguish cytokine factor receptor signaling, in which SOCS1 and JAK/STAT pathway are closely related. INF-γ、IL-10、IL-6 and other cytokines respectively through STAT3、STAT4、STAT6 of JAK/STAT signaling pathway induce the differentiation of Thl,Th17, Th2 cells, and then the Th1, Th17, Th2 cells secrete IL-2、IFN-γ、TNF. IL-17 and other cytokines, leading to the occurrence and development of inflammatory disease. SOCS1 can inhibit the JAK tyrosine kinase activity directly by binding with the kinase inhibitory region (KIR) of JAK kinase, thereby inhibiting the JAK/STAT signaling pathway. Studies have indicated that STAT3 and SOCS1 in the skin lesions of patients with psoriasis are higher than those of healthy people, which indicates that SOCS1 and STAT3 may be involved in the pathogenesis of psoriasis.Objective:To observe the effect of paeoniflorin and paeonol on HaCaT cells in psoriasis model to regulate the expression level of SOCS1, IL-1α, IL-1β, IL-6, IL-8.Methods:Prepare different concentrations of paeoniflorin, paeonol monomer and tripterygium glycosides containing serum. Intervene the HaCaT cells for 2 hours with each group containing serum,add a mixture of IL-6 and 50NG/ml 100ng/ml IFN-the 1.5ml stimulation in HaCaT cells, establish the activation of JAK/STAT signaling pathway of psoriasis cytological model. The RNA expression levels of IL-1, IL-1, IL-6, IL-8, SOCS1 were detected by real-time PCR method.Result:(1)The RNAexpression levels of IL-1, IL-1, IL-6, IL-8, SOCSlof model group and blank groupare different (P<0.05), suggesting that the model was successful;(2) Paeoniflorin 2.5mg/kg/D group can be up-regulated in HaCaT cells psoriasis model of SOCS1 gene expression level (P< 0.01); paeoniflorin 2.5mg/kg/ D group and paeoniflorin 5mg/kg/D group can cut HaCaT cells in psoriasis model IL-la and IL-1β and IL-6 gene expression level (P< 0.05), paeoniflorin 5mg/kg/D group can down-regulate the HaCaT cells in psoriasis model of IL-8 gene expression level (P< 0.05);(3)The concentration of paeonol groups of SOCS1, IL-6 gene expression level of no significant difference as compared with the model group (P> 0.05), paeonol 38.6mg/kg/d group, paeonol 77.4mg/kg/d group, paeonol 154.8mg/kg/d group can cut HaCaT cells in psoriasis model IL-1α,IL-1βgene expression level (P< 0.05), the groups of different concentrations of paeonol can cut HaCaT cells in psoriasis model of IL-8 gene expression level (P< 0.05).Conclusion:1. Paeoniflorin can up-regulatethe SOCS1 gene expression level and down-regulate the expression level of IL-la, IL-6, IL-1β,IL-8,and the effect ofhigh concentration group to inflammatory factors was more significant.2. Paeonol has inhibitory effect on HaCaT cellof psoriasis model of inflammatory factors, but it has no significant effect on SOCS1 gene.
