Recently, the number of male patients suffering from infertility shows the trend of incidence on the rise in the world. With a long history of treatment of male infertility, Traditional Chinese Medicine obtain a good curative effect and still a conventional therapy in the clinical. According to the Traditional Chinese Medicine, there is a hidden essence of Kidney and presides over reproduction which decides the male fertility. Kidney essence deficiency is a major cause of male infertility in clinical. Wuziyanzong pill(WYP), a classical formula in Chinese Medicine, is widely utilized for the treatment of male infertility with kidney essence deficiency. But the overall effectiveness and the mechanism is still unclear, therefore, research the mechanisms of WYP to better guidance for clinical medication is an urgent need.This thesis consists of two sections-literature review and experimental study. Literature review systematically summarize s the current research on infertility, summarizes the present situation of modern medical treatment of male infertility. It also reviewes the clinical and experimental research of WYP in recent years, The function of Wuziyanzong Pills (WYP) and Sertoli cell mitophagy on spermatogenesis was discussed.The experimental study mainly includes the following 4 aspects.Objective:To investigated the function of Wuziyanzong Pills (WYP) and Sertoli cell mitophagy on spermatogenesis.Methods:Five groups of male Sprague-Dawley rats including the control group, the model group, the low-dose WYP group, the medium-dose WYP group and the high-dose WYP group (10 in each group) were treated daily with vehicle, Tripterygium Wilfordii glycosides (GTW) either alone(20 mg/kg) or followed by WYP(0.5,1.0,or 2.0 g/kg daily), respectively for 30 days. The sperm motility, concentration and sperm count in the cauda epididymis were assessed by computer assisted sperm analysis. Methods Immunohistochemistry were employed to detect the expression of LAMP-2 in Sertoli cell. Examined the autophagosomes with an H-7100 transmission electron microscope.Results:The toxicity of GTW towards Sertoli cell and spermatogenesis was accompanied by decreased LC3 and LAMP-2 expression in the GTW group compared with the control group. The decreased quantification of LC3 immunogold particles in Sertoli cell were observed in the GTW group testis tissue. Oral high-dose WYP administrations to GTW-treated rats effectively increased all of the GTW-induced changes on autophagy in Sertoli cells. Furthermore, semen quality increased greatly compared with the model group.Conclusion:WYP has the ability to improve the spermatogenesis, possibly through modulating the autophagy of Sertoli cells.
