Study On The Expression Of Profilin-1 And Cofilin-1 Protein In SHR Renin-angiotensin-aldosterone And Aortic Wall

Objetives:This research is designed to clarify the function of empirical prescription by means of clearing liver and eliminating dampness in lowering blood pressure and comparethe levels of ALD、P&A、Ang In Ang Ⅱ from different groups determined by ELISAto investigate the mechanism of the treatment:which might be by inhibiting the renin-angiotensin-aldosterone system. And the experiment compares the levels of profilin-1 and cofilin-1 from different groups to clarify the function of empirical prescription on vascular remodeling because of hypertension.Methods:Pick up 50 SHR and 10 WKY that grow 13 weeks, which are male and female. All SHR are divided randomly in five groups:Captopril group (group Cap), High dosage Chinese medicine group (group High), Middle dosage Chinese medicine group (group Middle), Low dosage Chinese medicine group (group Low) and Control group (group Control), and 10 WKY are Normal group (group Model), which are male and female. Feed group Cap、High、 Middle and Low with corresponding drug dose and group Control and Model with water, 1ml/100g once a day last for four weeks. Measure all the rats’ tail artery pressure every week. After the forth week, all rats are performed carotid artery intubation to systolic blood pressure and drawn blood for serum preparation. By ELISA method, contents of ALD、PRA、Ang I、Ang Ⅱin serum are detected. After drawning blood, remove the thoracic aorta rapidly in liquid nitrogen for preserving to detect the levels of profilin-1 and cofilin-1 by Western Blot.Results:1. The effect on blood pressure in SHR by empirical prescription(1) the comparison of the weekly data on blood pressure:The blood pressure of group Model is significantly higher than group Control, and there is extremely significant difference (P<0.01). Compared with group Model, the blood pressure in group Cap and Middle are and lower since the first week(P<0.01), the blood pressure in group High and Low are and lower since the second week(P<0.01).(2) the comparison of carotid artery blood pressure:The blood pressure of group Model is significantly higher than group Control, and there is extremely significant difference (P<0.01). Compared with group Model, not only the blood pressure in group Cap is lower (P<0.01), but also the both systolic blood pressure in group Middle are lower(P<0.01), and diastolic blood pressure(P<0.05).2. The effect on RAAS in SHR by empirical prescription(1) ALD:The level of ALD in group Model is significantly higher than group Control, and there is extremely significant difference (P<0.01). Compared with group Model, not only the group Cap and Middle is lower (P<0.01), but also the group High and Low(P<0.05).(2) PRA:The level of PRA in group Model is significantly higher than group Control, and there is extremely significant difference (P<0.01). Compared with group Model, not only the group Cap and Middle is lower (P<0.01), but also the group High and Low(P<0.05).(3) Ang Ⅰ:The level of Ang Ⅰ in group Model is significantly higher than group Control, and there is extremely significant difference (P<0.01). Compared with group Model, not only the group Cap and Middle is lower (P<0.01), but also the group High and Low(P<0.05).(4) Ang Ⅱ:The level of Ang Ⅱ in group Model is significantly higher than group Control, and there is extremely significant difference (P<0.01). Compared with group Model, not only the group Cap and Middle is lower (P<0.01), but also the group High and Low(P<0.05).3. The effect on vascular remodeling by empirical prescription (1) profilin-1:The level of profilin-1 in group Model is significantly higher than group Control. Compared with group Model, all the drug groups are lower, and the result is:group Cap≈group Middle>group High≈group Low.(2)cofilin-1:The level of cofilin-1 in group Model is significantly higher than group Control. Compared with group Model, all the drug groups are lower, and the result is:group Cap≈ group Middle>group High≈group Low.Conclusion:1.Empirical prescription by means of clearing liver and eliminating dampness can reduce blood pressure in SHR and the effect is insistent and stable.2.Empirical prescription by means of clearing liver and eliminating dampness might lower blood pressure by inhibiting the renin-angiotensin-aldiotensin system.3.Empirical prescription by means of clearing liver and eliminating dampness could improve the situation of vascular remodeling because of hypertension.
