Abelmoschus manihot (L.) Medic.is an annual or perennial herb. As a kind of traditional Chinese medicine, it has a long history of application, and all parts of it can be used. However, the flower is its main medicinal part, the seed, root, stem and leaf are rarely used. Therefore, the resource of Abelmoschus manihothas not yet been fully utilized. Meanwhile, current researches on Abelmoschus manihot aremore concentrated on theflower, few studies involving its seed, root, stem and leaf. There is lack of comprehensive, systematic and comparative research on the different medicinal parts. And the quality evaluation standards of the related medicinal materials are not perfect. All of these have hindered the application and promotion of the medicinal plant.In this study, to solve the problems above, we conducted systematic and comparative experiments on the flower, seed, root, stem and leaf of Abelmoschus manihot from herbal textual research, content determination of total compositions (total flavonoids and total polysaccharides) and monomer composition, TLC identification. The specific experimental methods and results are as follows:1 Herbal textual researchReferring to the relevant literatures and traditional herbal works, we identified the scientific name is Abelmoschus manihot (L.) Medic, and five parts(the flower, seed, root, stem and leaf) of it can be used as medicine. The potency and efficacy of five medicinal parts are slightly different, but also have their similarities. They are sweet to taste, cold, have the effects of heat-clearing and detoxifying, inducing diuresis to alleviate edema.2 Extraction and content determination of total flavonoids in five medicinal parts of Abelmoschus manihot (L.) Medic.The chemical compositions of the flower of Abelmoschus manihot are mainly flavonoids. And the current researches on chemical constituents and pharmacological activities of Abelmoschus manihot are mainly concentrated on total flavonoidsand relevant monomer compositions of the flower. The extraction technology of total flavonoids in five medicinal parts of Abelmoschus manihot was respectively optimized by single factor and orthogonal experiment with the content of total flavonoids as the inspection index, using ethanol concentration, the ratio of material to solvent, extraction temperature and extraction time as factors. The results show that there are significant differences in the content of total flavonoids in different medicinal parts. The content of total flavonoids in the flower is far higher than that in other medicinal parts.3 Extraction and content determination of total polysaccharides in five medicinal parts of Abelmoschus manihot (L.) Medic.Experimental researches in recent years show that polysaccharide has many biological activities, and according to reports in the literature, the main chemical composition in the root, stem and leaf of Abelmoschus manihot is polysaccharide. In this experiment, by single factor and orthogonal experiment, we respectively optimized the extraction technology of total polysaccharides in five medicinal parts of Abelmoschus manihot, with water as extraction solvent, the content of total polysaccharides as the inspection index, using the ratio of material to solvent, extraction temperature and extraction time as factors. The results show that there are significant differences in the content of total polysaccharides in different medicinal parts.4 Content determination of Hyperoside in five medicinal parts of Abelmoschus manihot (L.) Medic.Recent studies show thatHyperoside has obvious pharmacological activities, and the content of which inthe flower of Abelmoschus manihotis high. In this study, we determined the content of Hyperoside in five medicinal parts of Abelmoschus manihot by HPLC. The results indicate: ①The content of Hyperoside in the flower is significantly higher than other medicinal parts, and there are large differences in the flowers of different regions; ②Part of Abelmoschus manihotleaf samples contain the composition, and the content is very low, most of the samples were not detected in the composition; ③The stem, root and seed of Abelmoschus manihotwere not detected in the composition.5 TLC identification of five medicinal parts of Abelmoschus manihot (L.) Medic.In this experiment, we selected the optimum polyamide thin layer chromatography conditions of the total flavonoids extracted from five medicinal parts of Abelmoschus manihot.Compared with other medicinal parts, the TLC of flower has more spots, and the spots are clear.
