Objectives:The aim of the study was to investigate the influence of Golli-MBP on calcium entry, apoptosis and proliferation abilities of T cells in vitro and to preliminarily establish animal models of OLP. Provide a basis for exploring the pathogenesis of immune-related diseases.Methods:Lentivirus mediated infection, qPCR and Western blot assay were performed to detect the mRNA and protein expression of Golli-MBP in Jurkat cells. Flow cytometry to detect relative intracellular calcium concentration and apoptosis. CCK-8 assay was performed to detect cell proliferation. Lentivirus was injected under Syrian golden hamsters’ oral mucosa, mucosal changes was detected by visual observation. Histological changes was detected by HE staining. Cell apoptosis was detected by TUNEL assay. Cell proliferation was detected by immunohistochemical method.Results:The mRNA and protein expression of lentivirus-infected Jurkat cells were over expressing. The apoptosis rate, proliferation rate and calcium influx of Golli-MBP-overexpressing Jurkat cells were decreased significantly. Golli-MBP failed to induce OLP lesions of experimental animals. There is no significant difference (p>0.05) of apoptosis rate in experimental groups and the control groups in the epithelia lamina on the 7th day and the apoptosis rate on 14th day in the experimental groups was lower than the control groups, the difference was statistically significant (p<0.05). Cortical cell proliferation rate on the 7th day and 14th day in the experimental groups was lower than the control groups and the difference was statistically significant (p<0.05).Conclusion:Over-expressing of Golli-MBP was able to inhibit Jurkat cells apoptosis and proliferation calcium influx ability significantly. Golli-MBP overexpression can regulate the proliferation and apoptosis of Syrian golden hamster cheek pouch mucosal epithelial cells. However, we failed to establish OLP animal models.
