Objective: To investigate and evaluate the effect of Tian Ma Tong Xin Nao by using the experimental rat model, and to explore the mechanism of improving the coronary artery atherosclerosis. Methods: the experimental rats were randomly divided into normal control group, model control group, positive control group(aspirin), high dose group and low dose group. Using pharmacological methods of Tian Ma Tong Xin Nao and on experimental atherosclerosis in rat blood rheology study index changes, by enzyme linked immunosorbent assay(ELISA) to determine the effects of serum IL-6 and TNF- α and observe the effect of the experimental atherosclerotic rat thoracic aorta and cardiac pathological changes. Results: The experimental results show that the each group of Tian Ma Tong Xin Nao could significantly inhibit the coronary atherosclerosis in rats model of red blood cell pressure product, degree of whole blood viscosity, plasma viscosity cell aggregation index, and erythrocyte deformation index of hemorheology index rise,the difference has statistical significance(P<0.01). The Tian Ma Tong Xin Nao cumulative concentrations of all the cardiac and cerebral side can significantly decrease the levels of IL-6 and TNF-α in the serum of the model rats with coronary atherosclerosis. The difference is statistically significant(P < 0.05 or P < 0.01). In H-E staining, the model group of rats in the thoracic aorta can see the local intimal thickening, uneven, vascular walls appear plaque, vascular lumen stenosis.Vascular endothelial cell proliferation in the vascular wall and a lesser raised plaque formation. was seen in the low dose group of Tian Ma Tong Xin Nao.And The high dose group showed vascular endothelial cell proliferation, intimal thickening was not smooth, there was no obvious plaque on the inner wall of the vessel. In the model group, the coronary artery of the heart of the model group was obviously thickened,not smooth, even the plaque of the inner wall of the vessel. A small amount of necrosis and lipid into atheroma, gather together into smaller patches uplift. Disorder of cardiac muscle fibers around the lesion in the low dose group of Tian Ma Tong Xin Nao.High dose treatment group of high dose treatment group showed vascular endothelial cell proliferation,intimal thickening was not smooth,there was no obvious plaque on the inner wall of the vessel.Conclusion:1. The Tian Ma Tong Xin Nao High dose group can improve the blood rheology of rat model with coronary artery atherosclerosis, and low dose effect is not obvious.2. The expression of TianMa TongXinNao High dose group could obviously inhibit the cardiovascular and coronary atherosclerosis rat model of serum IL-6 and TNF- α, and low dose effect is not obvious.3. The Tian Ma Tong Xin Nao High dose group can obviously slow down the process of blood vessel and heart in the rat model of coronary artery atherosclerosis, and low dose effect is not obvious.4. The TianMaTongXinNao in the treatment of cardiovascular and coronary atherosclerosis is related to inhibition of response to its good prediction,research direction and application prospects, and it was in a dose-dependent manner.
