Study On Acute Toxicity And Pharmacodynamics Of Mulberry Cough Granules

Purpose:Through the study on the pharmacodynamics and acute toxicity of Sangpi Zhike Granule,to investigate the anti-tussive and expectorant and anti-asthmatic and anti-inflamatory effects of this Granule and its acute toxic effect,provide the experimental basis for the clinical medication.Material and method:The experiment of relieving cough by ammonia induced cough in mice was observed and recorded the mice cough latency and the number of cough in 3min to evaluate the effect of relieving cough.Expectorant experiment using mouse phenol red expectoration method, measuring the absorbance(OD) of mouse tracheal phenol red excretion to evaluate the antitussive expectorant effects of Sangpi Zhike Granule.The anti-inflammatory test was used to evaluate the anti-inflammatory effect of the mouse ear swelling method, weighing and calculating the ear swelling degree of the mice.Antiasthmatic test with ovalbumin induced asthma in guinea pig, to observe and record the delitescence, evaluate the antiasthmatic effect.The acute toxicity test was administered by the maximum drug loading test(the maximum drug concentration and the maximum drug administration), 3 times in 24 hours, and 1 times every 8 hours.After 14 days, the general state of mice was observed and recorded. After the end of the observation, the weight and the quality changes of the mouse body were recorded. To dissect the mice and observe the abnormal phenomenon of the important organs with the naked eye.Results:Compared with the blank control group,Sangpi Zhike Granule low, medium,high dose group and milianchuanbei Pipagao group could prolong the incubation period of cough(P<0.05, P<0.01,) reduced cough frequency within 3min(P<0.05, P<0.01).Granule low, medium, high dose group and milianchuanbei Pipagao group absorbance(OD) value is significantly higher than the control group(P<0.05, P<0.01).Compared with the blank control group,the Swelling degree of angpi Zhike Granule low, medium,high dose group and aspirin group was significantly decreased.Sangpi Zhike Granule low, medium and high dose group and milianchuanbei Pipagao group compared with blank control group, the asthma latency was prolong(P<0.05, P<0.01).The mice were administered orally to the maximum dose of 612g·kg-1·d, equivalent to 360 times of clinical dosage of adults.Conclusion:1.Sangpi Zhike Granule has remarkable antitussive expectorant and anti-asthmatic effects and certain anti-inflammatory effect.2.Sangpi Zhike Granule has no obvious acute toxic effect on mice.
