Purpose:1. To observe the influences of the protein expressions of cardiac chloride channel CLC-2 and CLCA protein in acute myocardial ischemia mice induced by isoprenaline(ISO),and discuss the mechanism by along-channel acupuncture point to Neiguan(PC 6).2. Along-channel acupuncture point to Neiguan(PC 6) acupuncture was revealed has the specificity effect of acupuncture of the acute myocardial ischemia was induced by peritoneal injection of the ISO through the contrast research.Material and method:The ASIC3-/-knock out mice(n=37)for experiment were distributed randomly into 6 groups: control group(n=7), modeling control group(n=6), Neiguan(PC 6) treatment group(n=6), Lieque(LU 7) control group(n=6), Zusanli(ST 36) control group(n=6), and Non-acupoint control group(n=6). nonacupoint group at the point in middle of Tian Shu and Shen Que acupoints. The mice of all groups, except the control group, had myocardial ischemia by multiple subcutaneous injection of isoproterenol(ISO). Electroacupuncture treatment was given to Neiguan(PC 6) treatment group, Lieque(LU 7)control group, Zusanli(ST 36) control group and Non-acupoints control group, respectively, once a day for 7 days. After that,The S-T segment of the Ⅱ-lead ECG was recorded, the blood samples of the mice in each group were collected and marked with Eyeball vein Blood Collection Method, and their heart tissues were taken for back up. The changes of myocardial histology were observed by using microscope after HE staining.The content of superoxide dismutase(SOD)was detected by Colorimetric method, protein expression of CLC-2 and CLCA were detected by using Western blot method.Results:1.Compared with the S-T segment of the Ⅱ-lead ECG of mice injection of the ISO before and after, there is significance(p﹤0.05).2. In control group, myocardial fibers are in an ordered arrangement with morphological integrity, full nucleus, and clear boundary, which are visible 400 times Digital microscope.The HE staining sections of myocardial tissue showed that the sarcoplasm swelling,and necrosis area present weak staining in the modeling control group, the Lieque(LU 7) control group, the Zusanli(ST 36)group and the nonacupoint control group, cardiac tissue was observed with the assistance of HE staining; Compared with modeling control group, the sarcoplasm swellin and eosinophilic were relieved in Neiguan group(PC 6) group under the same physical microscope.3.Compared with the blank control group, the serum SOD levels of the modeling control group, the Lieque(LU7) control group, the Zusanli(ST 36)group and the non-acupoint control group were significantly lower(p< 0.01). Comparison with the modeling control group showed that the levels of serum SOD were also significantly increased in the Neiguan(PC 6) treatment group(p<0.01). No differences were found between the modeling control group, the Lieque(LU 7) control group, Zusanli(ST 36)group, and the nonacupoint control group(p>0.05). Compared with Neiguan(PC 6) group, SOD content decreased significantly(p<0.01)of the mice of Zusanli(ST 36)group, there is significant significance.4. Compared with blank control group, there was a significant increase in the contents of CLC-2, CLCA CL-channel protein in the modeling control group, the Lieque(LU-7) control group, Zusanli(ST 36)group, and the nonacupoint control group(p< 0.01).Comparison with the modeling control group showed that the contents of CLC-2,CLCA CL-channel protein was also significantly decreasd in the Neiguan(PC 6) treatment group(p< 0.01), the contents of CLC-2 CL-channel protein was also significantly decreasd in the Zusanli(ST 36)group(p< 0.01), the contents of CLCA CL-channel protein no differences were found in the Zusanli(ST 36)group(p﹥0.05).No differences were found between the modeling control group, the Lieque(LU 7) control group, and the nonacupoint control group(p>0.05). Compared with Neiguan(PC 6) group,the expressions of CLC-2 and CLCA significantly there is no significance(p﹥0.05)in the mice of Zusanli(ST 36)group.Conclusion:1. acupuncture at Neiguan(PC6) on mice with myocardial ischemia reflects the treatment efficacy of acupuncturing along channels. One of its treatment mechanisms can be interpreted by its adjustment of the protein expression of chloride channels CLC-2 and CLCA in myocardial cells, as proved in this study.2. The index is differ along-channel acupuncture the Neiguan(PC 6) and Zusanli(ST 36),it needs further research. Comparison study found that along-channel acupuncture the Neiguan(PC 6) to treatment of acute myocardial ischemia is specificity.
