| Objective:By observing the pathological changes of myocardial tissue,the content of serum CK-MB and the apoptosis of myocardial cells in rats with myocardial ischemia reperfusion,the effects of electroacupuncture at Xinshu and Neiguan points on prevention and treatment of myocardial ischemia-reperfusion injury were investigated.Methods:In this experiment,80 male Wistar rats were randomly divided into two groups: blank group,model group,ischemic preconditioning group and electroacupuncture pretreatment group.Myocardial tissues at each group immediately after reperfusion,6h,24 h,48h were observed.Blank group:before sampling,no factors were applied except normal binding feeding and the whole observation process.Model group:Before the model was established,the same group as the blank group was used.The left anterior descending artery of the coronary artery was ligated for 40 min and reperfused for 60 min.Electroacupuncture Pretreatment group: starting from the 15 th day before film formation,once a day,each time 30 min,a total of 15 days,5 days,rest for 1 day.Ischemic preconditioning group: Before the coronary artery ligation,the blood flow was blocked for 5 min,and then perfused for 5 min.This was repeated three times for 30 min of ischemic preconditioning.After the end of reperfusion,the rats in each group were subjected to blood collection in the abdominal aorta at the scheduled time.After the blood collection was completed,the rats were sacrificed and the samples were quickly taken.The pathological changes of rat myocardium were observed by HE staining.The serum CK-Mb content was detected by enzyme-linked immunosorbent assay(ELISA).The apoptosis of rat cardiomyocytes was detected by TUNEL method.Results:There were no obvious pathological changes in the blank group.The CK-Mb value of the model group was significantly increased at each time point.The pathological changes of myocardial tissue were observed by HE staining,and the apoptotic index of cardiomyocytes was significantly increased.Compared with the model group,the CK-Mb was significantly decreased in the electroacupuncture pretreatment group(P<0.01),and the CK-Mb values in the electroacupuncture pretreatment group and the ischemic preconditioning group were lower at 24 hours and 48 hours.The latter(p < 0.05).However,there was no significant difference between the two groups immediately and 6hours(p=0.609,p=0.085,p>0.05).Compared with the model group,the pathological changes of myocardial tissue in the electroacupuncture pretreatment group were significantly reduced,and the myocardial structure damage was lighter than that in the ischemic preconditioning group.Compared with the model group,the apoptotic rate of cardiomyocytes in the electroacupuncture pretreatment group was significantly decreased(p<0.01,p<0.05).The apoptotic index of cardiomyocytes in the 24 hour group after acupuncture pretreatment was significantly lower than that in the 24 hour group after ischemic preconditioning(p<0.05).However,there was no significant difference between the two groups immediately and 6 hours(p=3.38,p=0.36,p>0.05).Conclusion:1.Electroacupuncture at Xinshu and Neiguan Pretreatment significantly inhibited the pathological changes of myocardial tissue in rats with myocardial ischemia and reperfusion.2.Electroacupuncture Xinshu and Neiguan Pretreatment can significantly reduce serum CK-MB content and myocardial apoptosis index in rats with myocardial ischemia-reperfusion,protect myocardial tissue and improve myocardial tissue damage in rats with myocardial ischemia reperfusion3.Compared with the ischemic preconditioning group,electroacupuncture Xinshu and Neiguan pretreatment were better than ischemic preconditioning for myocardial protection after ischemia-reperfusion for 24 h and 48 h.4.Acupuncture preconditioning is safe and effective in preventing myocardial ischemia-reperfusion injury,and can be used for prevention and treatment of reperfusion injury in patients with ischemic coronary heart disease. |
