C.fluminea is a kind of high-yield economic shellfish. Due to its delicious taste and rich nutrition, it had great potential for development and utilization. The current research on C.fluminea by domestic and foreign scholars mainly focus on the C.fluminea polysaccharides and C.fluminea glycoprotein, there were few reports on the C.fluminea bioactive peptides. This study focuses on the extraction, separation and purification and antioxidant activity of C.fluminea peptides.1. Optimization of enzymatic extracted C.fluminea peptides process technology.To use C.fluminea as raw materials, DPPH radical scavenging rate as the response value. Compared with other enzymes, trypsin has the best enzymatic hydrolysis ability. Enzymatic hydrolysis experiment was carried out by trypsin, optimized process parameters by response surface design on the basis of the single factor experiment. Optimization of the process parameters:the content of enzyme was 1600U/g, pH was 8, enzyme hydrolysis temperature was 38.8℃, enzymatic hydrolysis time was 6.5 h. Under this condition, the DPPH scavenging rate was 43.25+0.17%.2. Study on antioxidant activity and stability activity of the ultrafiltration separated C.fluminea peptides.By ultrafiltration to separate the C.fluminea peptides, the low molecular weight (<3KD) of C.fluminea polyp eptide showed strong antioxidant activity. The results of study on stability activity of low molecular weight C.fluminea peptides showed that: ① The temperature had little influence on the activity of low molecular weight C.fluminea peptides;② Repeated freeze-thaw cycles had little influence on the activity of low molecular weight C.fluminea peptides;③ Alkaline condition had great influence on the activity of low molecular weight C.fluminea peptides;④ Heavy metal ions(Cu2\Zn2+) had great influence on the activity of low molecular weight C.fluminea peptides, heavy metal ions were not conducive to keep the ability of low molecular weight C.fluminea peptides.3. Separation and purification of low molecular weight C.fluminea peptides.6 peaks were separated by Sephadex G-15 of low molecular weight C.fluminea peptides. The result showed that the highest activity was peak 4(CFPs-4), IC50 of various indicators was the smallest, the scavenging rate of DPPH radical was 0.45mg/mL, the scavenging rate of hydroxyl radical was 0.25mg/mL. Then, used macroporous resin DA201-C to separate and purify the peak 4(CFPs-4). There were 3 peaks isolated from the peak 4 (CFPs-4), and the peak 3(CFPs-4-3) showed the strongest antioxidant activity, the scavenging rate of DPPH radical was 0.36mg/mL, the scavenging rate of hydroxyl radical was 0.23mg/mL4. The protective effect of low molecular weight C.fluminea peptides on chemical liver injury.The low molecular weight C.fluminea was applied in chemical liver injury mice which was builted by CCL4. There was almost no effect on weight by intake the low molecular weight C.fluminea. The liver/body ratio was significantly reduced; the spleen/body ratio was significantly improved by intake the right amount of low molecular weight C.fluminea. The result showed that the activity AST and ALT in serum、SOD and GSH-Px in liver were significant improved by the high mount of low molecular weight C.fluminea intake. At the same time, it can effectively reduce the generation of MDA in the liver, the liver was protected.
